目的:利用高效毛细管电泳(HPCE)的分离分析优势,建立海南本地中药材高良姜黄酮类提取物的毛细管电泳特征图谱。方法:采用HPCE法分析,未涂敷石英毛细管柱(48 cm×50μm),缓冲溶液成分20 mmol/L硼砂,30 mmol/L十二烷基硫酸钠,体积分数10%的乙腈(pH=8.7),运行电压为20 kV,紫外检测波长为208 nm,测定了14批海南本地不同市县高良姜药材样品的HPCE图谱,包括分离条件的优化、电泳峰的指认与标定、相似度评价及方法稳定性、精密度、重现性测定等。结果:建立了由15个共有峰组成的海南高良姜黄酮类提取物的高效毛细管电泳特征图谱,并鉴定了其中6个主要特征峰。结果表明海南岛不同市县的高良姜药材中黄酮类成分及含量有一定差别,依据电泳图谱中特征峰的迁移时间和峰面积能有效的鉴别不同产地高良姜药材。结论:该方法建立的海南高良姜的高效毛细管电泳图谱,进样量少,样品处理简单,分离效率高,污染小,结果可靠,重复性好。
OBJECTIVE: To establish a capillary electrophoresis profile of the flavonoids of Alpinia officinarum from Hainan, using the advantages of separation and analysis of high performance capillary electrophoresis (HPCE). Methods: The HPCE method was used to analyze the effect of HPCE on the surface area of human breast cancer cells. The results showed that no capillary column (48 cm × 50 μm), 20 mmol / L borax buffer, 30 mmol / L sodium dodecyl sulfate and 10% ), Operating voltage of 20 kV, UV detection wavelength of 208 nm, measured 14 batches of Hainan local GA galangal samples from different cities and counties, including the separation of the optimization conditions, electrophoresis peak identification and calibration, similarity evaluation and methods Stability, precision, reproducibility determination and so on. Results: The high performance capillary electrophoresis profiles of the flavonoids of Alpinia officinarum from 15 common peaks were established and six main characteristic peaks were identified. The results showed that there were some differences in the content and content of flavonoids in galangal medicinal materials in different cities and counties of Hainan Island. Based on the migration time and peak area of the characteristic peaks in the electrophoretic spectrum, the galangal herbs in different producing areas could be effectively identified. Conclusion: The high performance capillary electrophoresis of Alpinia officinarum collected by this method has less sample injection, simple sample processing, high separation efficiency, less pollution, reliable results and good repeatability.
