目的:探讨参麦注射液对单侧输尿管梗阻(UUO)大鼠健侧肾脏形态和功能代偿的影响。方法:成年雌性SD大鼠54只,随机分为假手术组、模型组和参麦注射液治疗组。假手术组仅将输尿管游离但不结扎离断,模型组和参麦注射液治疗组行单侧输尿管结扎术。术后治疗组每天腹腔注射参麦注射液,假手术组与模型组每天腹腔注射等量生理盐水。术后第7、14、21天随机选取并处死各组中的6只大鼠,取血5 m L监测肾功能的变化,健侧肾脏行肾皮质微循环血流检测,并观察双侧肾脏形态学改变。结果:模型组大鼠健侧肾脏体积明显增大,肾小球系膜细胞增生并随梗阻时间延长逐渐加重,血肌酐、尿素氮在术后第14天偏离正常水平;与假手术组比较,参麦注射液治疗组健侧肾脏形态和肾功能指标差异无统计学意义;模型组和参麦注射液治疗组健侧肾脏皮质微循环血流显著增加。结论:参麦注射液和模型组通过加快血细胞的运动速度而增加肾脏皮质微循环血流,促进单侧输尿管梗阻大鼠健侧肾脏的功能代偿,同时参麦注射液可能通过抑制肾小球细胞增殖而延缓或阻止其形态代偿。
Objective: To investigate the effect of Shenmai injection on contralateral kidney morphology and function in unilateral ureteral obstruction (UUO) rats. Methods: 54 adult female Sprague-Dawley rats were randomly divided into sham operation group, model group and Shenmai injection group. In the sham-operation group, only the ureter was detached but not ligatured. The model group and Shenmai injection group were given unilateral ureteral ligation. The patients in the treatment group were given intraperitoneal injection of Shenmai injection daily, and the sham operation group and model group were given the same amount of normal saline intraperitoneally every day. On the 7th, 14th and 21st day after surgery, 6 rats in each group were randomly selected and sacrificed, and the changes of renal function were monitored by 5 ml of blood. Renal cortex microcirculation blood flow was detected in the contralateral kidney and bilateral kidneys Morphological changes. Results: The volume of contralateral kidney in model group increased significantly, the mesangial cells proliferated and gradually increased with the prolongation of obstruction time. Serum creatinine and urea nitrogen deviated from the normal level on the 14th day after operation. Compared with the sham operation group, Shenmai injection group had no significant difference in contralateral kidney morphology and renal function indexes; microcirculation blood flow in contralateral renal cortex of model group and Shenmai injection group increased significantly. Conclusion: Shenmai injection and model group can increase microcirculation flow of renal cortex by accelerating the velocity of blood cells and promote the function compensation of contralateral kidney in rats with unilateral ureteral obstruction. At the same time, Shenmai injection may inhibit glomeruli Cell proliferation delays or prevents its morphological compensation.
