目的:本实验旨在采用仪器分析的手段,测定重组人红细胞生成素(rHuEPO)的唾液酸化程度。方法:利用rHuEPO中唾液酸带负电荷的特性,通过高效毛细管电泳分离了唾液酸化程度不同的rHuEPO。结果:pH4.0的磷酸盐缓冲液可有效地涂渍毛细管壁,减少电渗流,提高分辨率。在优化的测定条件下共分出了6种糖基化形式。结论:建立的HPCE法能有效地反映rHuEPO的纯度、微观不均一性和体内生物学活性
Objective: This experiment aims to determine the sialylation of recombinant human erythropoietin (rHuEPO) using instrumental analysis. Methods: Based on the negative charge of sialic acid in rHuEPO, rHuEPO with different degree of sialylation was isolated by high performance capillary electrophoresis. Results: Phosphate buffer pH 4.0 effectively coated capillary walls, reducing electroosmotic flow and improving resolution. Under the optimized conditions, 6 kinds of glycosylated forms were separated. Conclusion: The established HPCE method can effectively reflect the purity of rHuEPO, microscopic heterogeneity and in vivo biological activity
