目的:观察参麦注射液(SMI)对加入IL-1β诱导下软骨细胞Bcl-2/Bax mRNA表达的影响,探讨其防治软骨退变的作用机制。方法:分离培养体外软骨细胞,随机分成3组:空白组、IL-1β诱导组和参麦组,空白组采用含10%胎牛血清的培养基培养,IL-1β诱导组采用含浓度为10ng/mL IL-1β的培养基培养,参麦组采用含浓度为10ng/mL IL-1β和10%参麦注射液的培养基培养,采用反转录/聚合酶链反应(RT-PCR)法检测参麦注射液对软骨细胞Bax和Bcl-2 mRNA的表达情况。结果:与空白组比较,IL-1β诱导组Bcl-2 mRNA表达明显减少(P<0.01),Bax mRNA表达增多(P<0.01),均有统计学意义;与IL-1β诱导组比较,参麦组Bcl-2 mR-NA表达明显增多(P<0.01),Bax mRNA表达减少(P<0.01)。结论:参麦注射液可能通过降低促凋亡基因BaxmRNA表达,增加凋亡抑制基因Bcl-2mRNA的表达,从而调节Bcl-2/Bax mRNA的比例,抑制软骨细胞凋亡。
Objective: To observe the effect of shenmai injection (SMI) on the expression of Bcl-2 / Bax mRNA in chondrocytes induced by adding IL-1β, and to explore its mechanism of prevention and treatment of cartilage degeneration. Methods: Chondrocytes were isolated and cultured in vitro and randomly divided into 3 groups: blank group, IL-1βinduction group and Shenmai group, blank group were cultured with 10% fetal bovine serum medium, IL-1βinduced group with concentration of 10ng / mL IL-1β, and the Shenmai group was cultured in a medium containing 10ng / mL IL-1β and 10% Shenmai injection. Results: Compared with the blank group, the expression of Bcl-2 mRNA was significantly decreased (P <0.01) and the expression of Bax mRNA was increased in IL-1βinduced group (P <0.01) The expression of Bcl-2 mR-NA in wheat increased significantly (P <0.01), while the expression of Bax mRNA decreased (P <0.01). Conclusion: Shenmai injection may inhibit the apoptosis of chondrocytes by decreasing the expression of Bax mRNA, increasing the expression of Bcl-2 mRNA, and regulating the ratio of Bcl-2 / Bax mRNA.
