目的 :探讨雌激素对SD大鼠脂质代谢及血糖的影响。方法 :将40只7周龄的雌性SD大鼠随机分为4组:1对照组(Con组);2假手术组(sham组);3去卵巢组(OVX组);4去卵巢+雌激素替代组(OVX+E2组)。术后定期记录体重,给予相应处理,20周龄时测量空腹血糖并进行糖耐量试验,心脏穿刺取血,检测血浆总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C),同时取腹部脂肪称重。结果:与对照组相比,OVX组大鼠体重明显增加,腹部脂肪重量增加,血浆中TC、TG和LDL-C含量明显增加,空腹血糖无异常改变,但存在一定程度的糖耐量异常。OVX+E2组使用剂量雌激素替代治疗后,与对照组相比,体重明显减轻,腹部脂肪重量减轻,血脂无异常改变。结论:雌激素水平与体重呈负相关,雌激素水平下降可引起大鼠脂质代谢异常及胰岛素抵抗,补充雌激素后上述异常得到改善。
Objective: To investigate the effect of estrogen on lipid metabolism and blood glucose in SD rats. Methods: Forty seven-week-old female Sprague-Dawley rats were randomly divided into 4 groups: 1 control group (Con group), 2 sham group (sham group), 3 ovariectomized group (OVX group), 4 ovariectomized + Hormone replacement group (OVX + E2 group). The body weight was recorded regularly and the fasting blood glucose was measured at the age of 20 weeks. The blood glucose was measured by cardiac puncture and the contents of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL) -C) and high-density lipoprotein cholesterol (HDL-C), while taking abdominal fat weighing. Results: Compared with the control group, the body weight of OVX rats increased significantly, the weight of abdominal fat increased, the content of TC, TG and LDL-C in plasma increased obviously, and the fasting blood glucose did not change abnormally, but there was a certain degree of impaired glucose tolerance. Compared with the control group, the body weight of the OVX + E2 group was significantly reduced after the dose of estrogen replacement therapy was administered, the weight of abdominal fat was reduced, and the blood lipids did not change abnormally. Conclusion: The level of estrogen is negatively correlated with body weight. The decline of estrogen level may cause lipid metabolism and insulin resistance in rats, and the above abnormalities are improved after estrogen replacement.
