采用直线加速器照射制作动物的β损伤模型,同时对ΝΙΗ 3Τ3细胞进行β外照射,观察胶原总量及Ⅰ,Ⅲ型胶原含量的改变;观察胶原降解酶ΜΜPs-1活性变化;同时测定损伤后细胞因子ΤGF-β1,IL-6的变化情况。结果表明,β损伤后胶原总量变化不大,而Ⅰ型胶原含量下降,Ⅲ型胶原含量上升;MMPs-1 活性上升;β损伤后TGF-β1,IL-6 表达量增加。提示胶原代谢的变化在放射性皮肤损伤中起关键作用,损伤后胶原代谢的调控机制中,TGF-β1 和 IL-6 起重要作用。
The model of β-injury was made by linear accelerator. At the same time, β-irradiation was performed on ΝΙΗ 3T3 cells to observe the change of total collagen and collagen type I and III. The activity of collagen degrading enzyme ΜΜPs-1 was observed. Factors of TGF-β1, IL-6 changes. The results showed that the total amount of collagen did not change much after β-injury, but the content of type Ⅰ collagen decreased, the content of type Ⅲ collagen increased, the activity of MMPs-1 increased and the expression of TGF-β1 and IL-6 increased after β-injury. It is suggested that the changes of collagen metabolism plays a key role in the radioactive skin injury. TGF-β1 and IL-6 play an important role in the regulation of collagen metabolism after injury.
