目的 :研究全反式维甲酸 (ATRA)对晶体上皮细胞 (LECs)增殖的抑制作用。方法 :取体外培养的第 2代兔晶体上皮细胞 (RLECs) ,分为正常培养 (对照组 )和加入不同浓度 (1× 10 - 8~ 1× 10 - 5mol/L)ATRA的实验组 ,分别于培养后 2 4h和 4 8h观察各组晶体上皮细胞的形态学改变 ,计算各浓度ATRA的增殖抑制率。结果 :各种浓度的ATRA对兔晶体上皮细胞均有明显的抑制作用。随浓度升高 ,细胞形态异常改变趋于明显 ;其抑制率由低浓度时的 14 .0 %增至高浓度时的 75 .7% ;1× 10 - 6 mol/L为最高有效浓度 ,半效量 (LD50 )浓度为 0 .85× 10 - 6 mol/L。结论 :ATRA对体外培养的兔晶体上皮细胞增殖有明显抑制作用
Objective: To study the inhibitory effect of all-trans retinoic acid (ATRA) on the proliferation of human epithelial cells (LECs). Methods: The second generation of rabbit lens epithelial cells (RLECs) were cultured in vitro and divided into experimental group (control group) and ATRA group (1 × 10 -8 ~ 1 × 10 -5 mol / L) Morphological changes of lens epithelial cells were observed 24 h and 48 h after culture, and the proliferation inhibition rate of ATRA at each concentration was calculated. Results: Various concentrations of ATRA significantly inhibited rabbit lens epithelial cells. With the increase of concentration, the change of cell morphology tended to be obvious; the inhibition rate increased from 14.0% at low concentration to 75.7% at high concentration; 1 × 10-6 mol / L was the highest effective concentration and half effective The concentration of LD50 was 0.85 × 10 -6 mol / L. Conclusion: ATRA can significantly inhibit the proliferation of rabbit lens epithelial cells cultured in vitro
