目的 为了解海南省麻疹暴发病例中,现行麻疹野病毒流行状况,以及本土麻疹病毒的基因型。方法来集麻疹疑似病例咽试子标本,应用B95a细胞进行病毒分离培养。结果 在5例疑似病例中分离到2株麻疹病毒,阳性率为40%。经 RT-PCR和其产物的基因序列分析,属H1基因型,为中国本土麻疹野毒株。同时采用麻疹疫苗初免儿童血清中和新分离麻疹病毒,结果中和抗体滴度为1:10、1:20,说明我国生产的麻疹疫苗对我省现流行的麻疹野病毒能起到保护作用。结论 这是海南省首次用B95a细胞成功分离到麻疹野病毒,并通过基因分型证实为中国本土野病毒,填补了海南省麻疹病毒分离的空白。
Objective To understand the prevalence of measles virus in Hainan Province, the prevalence of the wild measles virus and the genotype of the local measles virus. Method to collect suspected cases of measles pharyngeal specimens, the use of B95a cells for virus isolation and culture. Results Two measles viruses were isolated from five suspected cases, the positive rate was 40%. The gene sequence analysis of RT-PCR and its products belongs to H1 genotype, which is a wild type measles virus in China. At the same time, measles vaccine was used to immunize the newly isolated children’s serum and newly isolated measles virus. The results showed that the titer of neutralizing antibody was 1:10,1:20, indicating that the measles vaccine produced in our country could protect the wild measles virus . Conclusion This is the first time that Hainan Province has successfully isolated the measles wild virus with B95a cells and confirmed by genotyping as the wild-type domestic virus in China, filling in the blank of measles virus isolation in Hainan Province.
