目的:研究子痫前期患者母血及脐血中AnnexinⅤ、ET-1的表达水平及与胎儿生长受限相关性,并探讨子痫前期发病机理及其与胎儿生长受限的关系。方法:采用ELISA方法检测子痫前期患者84例、FGR患者24例及对照组29例患者母血及脐血中AnnexinⅤ、ET-1的表达水平。结果:1子痫前期各组母血、脐血中annexinⅤ的蛋白表达水平均低于对照组,各组分别比较,差异均有高度统计学意义(P<0.01)。在母血及脐血中均是早发组<晚发组<轻度组,但母血中晚发组与轻度组比较,差异无统计学意义(P>0.05)。余各组两两分别比较,差异均有高度统计学意义(P<0.01)。各组与FGR组比较,FGR组母血及脐血中annexinⅤ的表达水平均低于轻度组、晚发重度组、对照组,分别与各组比较,差异均有统计学意义(P<0.01)。但FGR组与早发重度组比较,差异无统计学意义(P>0.05)。2各组孕妇母血及脐血中ET-1水平变化,子痫前期各组均明显高于对照组,分别比较,差异均有高度统计学意义(P<0.01)。在母血中早发重度型>晚发重度型>轻度子痫前期患者,差异仍有统计学意义(P<0.05)。但在脐血中轻度组与重度组比较,差异均有高度统计学意义(P<0.01);但早发组与晚发组比较,差异无统计学意义(P>0.05)。各组与FGR组比较,FGR组ET-1表达高于对照组及轻度组,且分别比较差异有高度统计学意义(P<0.01)。但早发型>FGR组>晚发型,但分别比较,差异均无统计学意义(P>0.05)。3AnnexinⅤ子痫前期各组及FGR组在孕妇外周血与脐血中的表达均呈明显正相关(P<0.01);ET-1子痫前期各组及FGR组在孕妇外周血与脐血中的表达均呈明显正相关(P<0.01)。结论:患者母血及脐血中AnnexinⅤ、ET-1的表达可能与子痫前期发病机制有关,通过影响胎盘中凝血-纤溶系统及血管内皮系统,参与子痫前期及胎儿生长受限发生、发展的病理生理过程。
Objective: To investigate the expression of AnnexinⅤ and ET-1 in maternal blood and umbilical cord blood of patients with preeclampsia and its relationship with fetal growth restriction, and to explore the pathogenesis of preeclampsia and its relationship with fetal growth restriction. Methods: The expression of Annexin V and ET-1 in maternal blood and umbilical cord from 84 patients with preeclampsia, 24 patients with FGR and 29 controls were detected by ELISA. Results: 1 The protein expression levels of annexin V in maternal blood and umbilical cord blood in preeclampsia group were lower than those in control group. There was a significant difference between each group (P <0.01). In both maternal and umbilical blood, they were early onset group 0.05). Each group was compared with each other, the differences were highly statistically significant (P <0.01). Compared with FGR group, the expression levels of annexin V in maternal blood and umbilical cord blood in FGR group were lower than those in mild group, severe late-onset group and control group, respectively (all P <0.01) ). However, there was no significant difference between FGR group and early-onset severe group (P> 0.05). The levels of ET-1 in maternal blood and umbilical cord blood in each group were significantly higher than those in the control group (P <0.01). The differences were statistically significant (P <0.01). In maternal blood, the patients with early-onset severe type> late-onset severe type> mild preeclampsia still had significant difference (P <0.05). However, there was a significant difference between the mild group and the severe group in cord blood (P <0.01), but there was no significant difference between early onset group and late onset group (P> 0.05). Compared with FGR group, the expression of ET-1 in FGR group was higher than that in control group and mild group (P <0.01). But early hair> FGR group> late-onset type, but there was no significant difference between them (P> 0.05). The expression of 3AnnexinⅤ in preeclampsia group and FGR group was positively correlated with umbilical cord blood in pregnant women (P <0.01). In ET-1 preeclampsia group and FGR group, Expression were significantly positive correlation (P <0.01). Conclusion: The expression of Annexin V and ET-1 in maternal blood and umbilical cord blood may be related to the pathogenesis of preeclampsia. It may participate in the development of preeclampsia and fetal growth by affecting the coagulation-fibrinolytic system and vascular endothelial system in the placenta. Development of the pathophysiological process.
