目的通过检测乳腺癌特异基因(breast cancer specific gene 1,BCSG1)在乳腺癌的表达,分析其与乳腺癌各项临床病理参数的相关性,评估BCSG1mRNA表达对乳腺癌预后的判断作用。方法随机抽取上海交通大学医学院附属新华医院1999年9月至2003年5月收治的93例女性乳腺癌病人的临床资料。采用逆转录-链式聚合酶反应(reverse transcription-polymerase chain reaction,RT-PCR)检测标本BCSG1 mRNA表达,统计分析BC-SG1基因表达与乳腺癌各项临床病理参数的相关性;跟踪随访93例乳腺癌病例,随访期为11~75个月,做生存分析,确立影响乳腺癌预后的独立因素。结果BCSG1在乳腺癌中表达率为36.6%(34/93),其中Ⅲ/Ⅳ期乳腺癌占76.5%(26/34)。BCSG1基因表达与乳腺癌肿瘤分期(P=0.000)和淋巴结转移状况(P=0.036)正相关。BCSG1表达阳性病例中位生存期为67.5个月,3年和5年无病存活率分别为78.8%和36.3%,明显低于BCSG1表达阴性病例(P=0.043)。BCSG1(P=0.039)和PR(P=0.038)为乳腺癌的独立预后因素。结论BCSG1基因表达与乳腺癌的肿瘤分期和淋巴结转移状况正相关;BCSG1可能是乳腺癌一种新的独立预后指标。
Objective To detect the expression of breast cancer specific gene 1 (BCSG1) in breast cancer and analyze its correlation with the clinicopathological parameters of breast cancer so as to evaluate the role of BCSG1 mRNA in the prognosis of breast cancer. Methods The clinical data of 93 female breast cancer patients who were admitted to Xinhua Hospital Affiliated to Shanghai Jiaotong University School of Medicine from September 1999 to May 2003 were randomly selected. The expression of BCSG1 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR), and the correlation between BC-SG1 gene expression and clinical pathological parameters was statistically analyzed. The follow-up of 93 cases Breast cancer cases, the follow-up period of 11 to 75 months, do the survival analysis, the establishment of an independent factor affecting the prognosis of breast cancer. Results The positive rate of BCSG1 in breast cancer was 36.6% (34/93), of which, stage Ⅲ / Ⅳ breast cancer accounted for 76.5% (26/34). BCSG1 gene expression was positively correlated with breast cancer staging (P = 0.000) and lymph node metastasis (P = 0.036). The median survival time was 67.5 months in BCSG1-positive cases, and 78.8% and 36.3% in 3 and 5 years respectively, which was significantly lower than that in BCSG1-negative cases (P = 0.043). BCSG1 (P = 0.039) and PR (P = 0.038) were independent prognostic factors for breast cancer. Conclusion BCSG1 gene expression is positively correlated with tumor stage and lymph node metastasis in breast cancer. BCSG1 may be a new independent prognostic factor in breast cancer.
