The design,synthesis,and biological activity evaluation of the potency of new inhibitors of sphingosine kinases 1 and 2(SphK1 and SphK2),the enzyme that catalyzes the phosphorylation of D-erythro-sphingosine to produce the key signaling lipid,sphingosine 1-phosphate1,are described.Recently,we reported that(E)-N-(4-(benzyloxy)benzylidene)-3,4,5-trihydroxybenzohydrazide(compound 25)was a novel inhibitor of SphK1 and SphK22.
