Surface plasmon resonance imaging (SPRi) [1-3] is a new, labeling-free and high-throughput analytical tool capable of imitating biological conditions and conducting batch researches.In this laboratory, SPRi has been explored since 1997 in order to conduct parallel investigation of biological reactions or related events.After design and fabrication of bifunctional SPR/SPRi equipments [1-2], a set of methods and/or protocols have been established for batch analysis of molecular recognition reactions [3-11].To further explore the advantages of SPRi, its hyphenation features have been studied recently with micro isoelectric focusing (μ-IEF), micro thin layer chromatography (μ-TLC), capillary electrophoresis (CE) and gas chromatography (GC) as targets.Some interesting features have been observed during these studies together with challenges.The most serious challenge is that SPRi can only "feel" a depth of <200 nn, better <50 nm.This is actually a type of near-field technique and all molecules needed to detect should be in close contact with the SPRi sensing film.
