Proteomics technology in combination with mass spectrometry analysis and database information analysis are the important ways to analyze and identify overall regulation target and signaling networks for TCM.TQEPD(Tonifying qi eliminating phlegm decoction) is an empirical prescription which has good clinical efficacy in treatment of non small cell lung cancer(NSCLC).The present study was aimed at reveal its mechanism in the treatment of NSCLC.To observe the effect of Yiqi Chutan Tang on Lewis lung cancer tumor growth in C57 BL mice and protein screening.Lung cancer tumor model was established in C57 BL mice.40 mice were randomly divided into model group and treatment group Tumor size,tumor weight and the numbers of lung metastases were detected.The expressed differentially protein spots information were acquired by a two-way fluorescence difference gel electrophoresis (2D-DIGE) system,and the differential proteins were identified by matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-TOF).We found that tumor size,tumor weight,tumor metastasis nodules in TQEPD treatment group were significantly less than that in model group.For gel diagram analysis of 2D-DIGE system,there were 44 expressed differentially protein spots,of which 37 proteins including 30 down-regulated proteins and 7 up-regulated proteins were successfully identified by MALDI-TOF-TOF in the TQEPD treatment group compared with model group.Among these down-regulated proteins,PGK,enolase 1,PGM5 were the rate-limiting enzyme of glycolysis.Hsp-60,P4HB,PDI-A3,Hsp5p,Hsp9,STIP1 were related to unfolded protein response which can be actived by glucose deprivation and hypoxia.All above proteins was highly expressed or activated in cancer cell to modulate cell metabolism in adapt to glucose deprivation.Cancer cells display high rates of aerobic glycolysis,a phenomenon known historically as the Warburg effect.Collectively,The possible mechanism of TQEPD in the treatment of LEWIS lung cancer was highly related with the down-regulated protein expression of,suggesting TQEPD may inhibit tumor growth and metastasis of lung cancer through altering cell metabolism.
