神经生长抑制因子(GIF)又名金属硫蛋白-Ⅲ, 是神经系统中第一个被鉴定的具有神经元生长抑制功能的蛋白. 采用MTT还原测定法测定了重组GIF对嗜铬细胞瘤株PC12和成神经细胞瘤株SY5Y生长的影响: 重组GIF在体外可抑制PC12的生长; 在浓度为100 mg/L时, 对PC12的抑制率约为25%; 而当浓度为300 mg/L时, 对PC12的抑制率达到约50%. 表明PC12可作为神经元模型来测定GIF的神经元生长抑制活性. GIF在体外不能抑制常用作神经细胞瘤株模型的SY5Y, 表明GIF并不具有广谱的抑制神经细胞瘤株的作用; GIF抑制PC12可能就是针对它所具有的胆碱能神经元的某些特性. 这对于揭示GIF的神经元生长抑制活性的作用机制有着重要意义.
