为探讨丙型肝炎病毒 (HCV)NS 5b区与 5’ -非编码区 ( 5’ -NCR)不同基因区RNA检测结果的区别及临床应用价值。利用逆转录聚合酶链反应 (RT -PCR)技术检测 148例肝炎患者血清HCVNS 5b基因的cDNA ,并与5’ -NCR基因区cDNA检测技术进行了比较。结果 HCVNS 5b基因区RNA检出率为 2 4.3% ( 34/ 148) ,5’ -NCR基因区RNA检出率为 31.1% ( 47/ 148)。HCV 5’ -NCR基因区扩增效果较好 ,而HCVNS 5bPCR敏感性接近于HCV 5’-NCR检测结果 ,亦可直接应用于临床RNA检测。
To investigate the difference and clinical value of RNA detection between different regions of HCV NS 5b and 5 ’non-coding region (5’ -NCR). The cDNA of serum HCV NS 5b gene in 148 cases of hepatitis patients was detected by reverse transcriptase-polymerase chain reaction (RT-PCR) and compared with that of 5 ’-NCR gene. Results The detection rate of RNA in HCV NS 5b gene region was 23.3% (34/148), and that of 5 ’-NCR gene region was 31.1% (47/148). HCV 5 ’-NCR gene region amplification effect is better, and HCVNS 5bPCR sensitivity close to HCV 5’-NCR test results can also be directly applied to clinical RNA detection.
