采用气管条浴槽实验方法 ,观察青心酮 (DHAP)对离体豚鼠气管螺旋条张力的影响。结果发现 :DHAP明显松弛豚鼠的离体气管平滑肌 ;抑制由乙酰胆碱 (Ach)、组胺 (His)和氯化钾所致离体豚鼠气管平滑肌的收缩反应 ;对 5 -羟色胺 (5 - HT)诱发的由内钙释放引起的 相收缩和外钙内流所致的 相气管平滑肌收缩均有一定的抑制作用。 3.96× 10 - 5 m ol/L DHAP则仅抑制 相收缩。随着其剂量的增大 (>10 - 4 mol/L ) , 相收缩也被抑制 ,上述抑制作用均呈剂量依赖性 ;消炎痛对 DHAP松驰离体气管平滑肌作用无明显影响。提示 DHAP这一效应可能与其钙拮抗有关 ,而与其抑制环加氧酶的作用关系则不甚密切
The effect of Qingxin Ketone (DHAP) on the tension of isolated rat guinea pig tracheal spiral strips was observed using a tracheal strip bath test. The results showed that: DHAP significantly relaxes the isolated trachea smooth muscles of guinea pigs; inhibits the contractile response of guinea pig tracheal smooth muscle induced by acetylcholine (Ach), histamine (His) and potassium chloride; induces 5-hydroxytryptamine (5-HT) The phase contraction caused by the release of calcium and the extracellular calcium influx caused by the contraction of the tracheal smooth muscle all have certain inhibitory effects. 3.96 × 10 -5 m ol/L DHAP only inhibited phase contraction. With the increase of its dose (>10 - 4 mol/L), the phase contraction was also inhibited. The above inhibitory effects were dose-dependent; indomethacin had no significant effect on DHAP relaxation in vitro tracheal smooth muscle. It is suggested that the effect of DHAP may be related to calcium antagonism but not to its role in inhibiting cyclooxygenase.
