目的探讨大鼠脑内注射凝血酶(Thrombin,TM)对细胞间粘附分子-1(Intracellular adhensionmolecule-1,ICAM-1)mRNA表达及白细胞浸润的影响及其可能机制。方法TM、TM+组织蛋白酶G(Ca-thepsin G,CATG)脑内立体定向注射于SD大鼠尾状核,在不同的时间点处死动物,RT-PCR技术检测ICAM-1mRNA表达,通过测定髓过氧化物酶(MPO)活性来检测中性粒细胞的浸润情况。结果凝血酶脑内注射后24h ICAM-1mRNA表达开始增加(P<0.05),48h达高峰(P<0.01),然后逐渐下降。MPO活性动态变化趋势与ICAM-1mRNA相似。CATG组ICAM-1mRNA及MPO活性与对照组比较差异均无显著性(P>0.05)。结论TM通过蛋白酶激活受体-1(protease activated receptor-1,PAR-1)诱导ICAM-1mRNA,促进白细胞浸润。
Objective To investigate the effects of thrombin (TM) on the expression of intracellular adhension molecule-1 (ICAM-1) mRNA and leukocyte infiltration in rat brain and its possible mechanism. Methods TM and TM + cathepsin G (CATG) were intracerebrally injected into the caudate nucleus of SD rats. Animals were sacrificed at different time points. The expression of ICAM-1 mRNA was detected by RT-PCR. Oxidase (MPO) activity to detect neutrophil infiltration. Results ICAM-1 mRNA expression began to increase (P <0.05) 24h after intracerebral injection of thrombin and peaked at 48h (P <0.01), then gradually decreased. The dynamic trend of MPO activity is similar to that of ICAM-1 mRNA. ICAM-1mRNA and MPO activity in CATG group were not significantly different from those in control group (P> 0.05). Conclusion TM induces ICAM-1 mRNA through protease activated receptor-1 (PAR-1) and promotes leukocyte infiltration.
