目的:建立高效液相色谱测定谷维素片含量的方法,并分析比较不同生产厂家生产的谷维素片的主要成分。方法:采用Agela Innoval C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-甲醇-二氯甲烷-冰醋酸(50∶50∶0.3∶0.3)为流动相,流速1.0ml·min~(-1),检测波长326 nm,柱温30℃,进样量20μl。检测国产与日产谷维素片主要成分。结果:谷维素在20.10~100.50μg·ml~(-1)范围内线性关系良好(r=1.000 0),平均回收率为101.2%(RSD=0.4%,n=9)。国产谷维素片主要成分相同,主要成分的含量存在差异,日产谷维素片与国产谷维素片比较,主要成分及其含量均存在较大差异。结论:该方法简便、准确可靠,既能用于谷维素片的含量测定,亦能用于谷维素片主要成分的分析比较。
Objective: To establish a HPLC method for the determination of oryzanol tablets and to analyze and compare the main components of oryzanol tablets produced by different manufacturers. Methods: The chromatographic separation was performed on a Agela Innoval C 18 column (250 mm × 4.6 mm, 5 μm) using acetonitrile-methanol-methylene chloride-glacial acetic acid (50:50:0.3:0.3) as the mobile phase at a flow rate of 1.0 ml · min ~ (-1), detection wavelength 326 nm, column temperature 30 ℃, injection volume 20μl. Detection of domestic and Nisshin oryzanol main components. Results: Oryzanol had good linearity (r = 1.000 0) in the range of 20.10-100.50μg · ml -1 with an average recovery of 101.2% (RSD = 0.4%, n = 9). The main components of the Chinese-made oryzanol tablets are the same with different contents of the main components. There are big differences between the main components and the content of the Chinese-made oryzanol tablets. Conclusion: The method is simple, accurate and reliable. It can be used for the determination of oryzanol tablets and the comparative analysis of the main components of the oryzanol tablets.
