目的探讨实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)非敏感品系Wistar大鼠加用不同剂量牛型结核杆菌卡介苗(mycobacterium bovis bacillus calmette-guerin,M.bovis BCG)对EAE模型诱导的作用。方法将Wistar大鼠分为EAE1、EAE2、EAE3、EAE4共4组,然后分别用含4、8、10、12 mg M.bovis BCG/mL完全福(氏)佐剂(CFA)加豚鼠脑脊髓匀浆(GPSCH)制备不同浓度的完全抗原,分别免疫诱导Wistar大鼠EAE。观察不同剂量M.bovis BCG对诱导EAE模型发病情况、各项临床指标的影响,同时取脑和脊髓制成石蜡切片,行HE染色观察组织形态变化,行单核细胞趋化蛋白-1(MCP-1)mRNA原位杂交(ISH)计数其阳性细胞百分率。结果EAE3、EAE4组的发病率、体重减轻、临床症状评分、MCP-1 mRNA阳性细胞数显著高于EAE1、EAE2组(均P<0.05)。结论M.bovis BCG浓度的高低影响Wistar大鼠EAE的发病率。
Objective To investigate the effects of different doses of mycobacterium bovis bacillus calmette-guerin (M. bovis BCG) on the induction of EAE in Wistar rats with experimental autoimmune encephalomyelitis (EAE) effect. Methods Wistar rats were divided into 4 groups: EAE1, EAE2, EAE3 and EAE4, and then treated with 4, 8, 10, 12 mg M.bovis BCG / mL complete Fufang adjuvant (CFA) Homogenate (GPSCH) to prepare different concentrations of the complete antigen, respectively, immune induction of Wistar rats EAE. The effects of different dosages of M. bovis BCG on the incidence of EAE model and the clinical indicators were observed. At the same time, the brain and spinal cord were made into paraffin sections. The morphological changes were observed by HE staining. The expression of monocyte chemoattractant protein-1 -1) mRNA in situ hybridization (ISH) count the percentage of positive cells. Results The incidence of EAE3 and EAE4 group was significantly higher than that of EAE1 and EAE2 group (all P <0.05). Conclusion The concentration of M. bovis BCG affects the incidence of EAE in Wistar rats.
