目的:研究化积方胶囊体内外抗肿瘤活性。方法:采用MTT法观察化积方对S180细胞生长抑制情况;鼠肝癌H22细胞株接种小鼠体内观察化积方对肿瘤抑制率和对小鼠的体质量抑制率;流式细胞仪定量检测其细胞周期和凋亡发生率的影响。结果:10,100,500μg.ml-1化积方对小鼠S180肿瘤细胞的增殖抑制率(%)分别为23.44,40.35,48.79(P<0.05),小鼠S180细胞的半数抑制浓度(IC50)为(0.554±0.027)mg.ml-1;5,15,45 mg.kg-1.d-1化积方均对H22细胞产生了抑制作用,抑制作用(%)分别为26.94,48.34,68.27(P<0.05)。10,100,500μg.ml-1的化积方给药组的细胞凋亡率分别为5.77%,12.03%和15.48%,均明显高于阴性对照组,两组差异有统计学意义(P<0.05)。结论:化积方胶囊具有抗肿瘤作用,这一作用可能与促进肿瘤细胞凋亡有密切联系。
Objective: To study the anti-tumor activity of Huji prescription square in vitro and in vivo. Methods: MTT method was used to observe the inhibitory effect of Huji Recipe on the growth of S180 cells. The inhibitory rate of Hu Ji Fang on the tumor inhibition rate and the body mass inhibition rate of mice were observed in vivo in H22 hepatoma cell inoculation mice. The expression of S180 was detected by flow cytometry Cell cycle and the incidence of apoptosis. Results: The inhibitory rates (%) of 10,100, 500μg.ml-1 formula product on S180 tumor cells were 23.44,40.35,48.79 (P <0.05) respectively. The IC50 of S180 cells was ( 0.554 ± 0.027) mg.ml-1, 5,15 and 45 mg.kg-1.d-1 all have inhibitory effect on H22 cells, the inhibitory effect (%) is 26.94,48.34,68.27 (P <0.05). The apoptotic rates of 10,100 and 500μg.ml-1 were 5.77%, 12.03% and 15.48%, respectively, which were significantly higher than that of the negative control group (P <0.05). Conclusion: Huji Jifang capsule has anti-tumor effect, which may be closely related to the promotion of tumor cell apoptosis.
