目的 :初步探讨马蔺子甲素体外抗鼻咽癌作用和诱导鼻咽癌细胞凋亡作用。方法 :细胞毒测定以MTT法 ,细胞凋亡的测定以流式细胞仪法。结果 :马蔺子甲素对多种鼻咽癌细胞株均具有细胞毒作用 ,对CNE_2、SUNE_1、Fadu细胞的IC50 分别为 1 8.8± 3.1、2 2 .4± 1 2 .0、1 9.3± 6 .8μmol L ;马蔺子甲素也能诱导鼻咽癌细胞株CNE_2细胞凋亡。Fadu细胞与 5 8.8、2 9.4μmol L浓度马蔺子甲素共同培养 48h ,其诱导细胞的凋亡率分别为 5 9.3%± 4.5 %、33.2 %±2 .3% ,半数凋亡浓度为 5 6 .3μmol L ,且具有剂量依赖性。结论 :马蔺子甲素本身也具有抗鼻咽癌作用及诱导鼻咽癌细胞凋亡的作用
Objective: To investigate the anti-nasopharyngeal carcinoma effect and induction of apoptosis in nasopharyngeal carcinoma cells in vitro. Methods: The cytotoxicity assay was performed by MTT assay and apoptosis was measured by flow cytometry. RESULTS: Horsetail anthraquinone had cytotoxicity on a variety of nasopharyngeal carcinoma cell lines, and the IC50 values on CNE_2, SUNE_1, and Fadu cells were 18.8±3.1, 22.4±12.0, and 19.3±6, respectively. 8μmol L; madridinone can also induce apoptosis in nasopharyngeal carcinoma cell line CNE_2. Fadu cells were co-cultured with 5 8.8 and 29.4 μmol L concentrations of madridinone for 48 h. The apoptotic rates of the induced cells were 5 9.3%±4.5% and 33.2%±2.3%, respectively. The half-apoptotic concentration was 5 6. 3 μmol L and dose-dependent. Conclusion : Horse melittin itself also has anti-nasopharyngeal carcinoma effect and induces apoptosis of nasopharyngeal carcinoma cells.
