目的:初步探讨人类组织激肽释放酶基因8(kallikrein gene8,KLK8)在乳腺疾病中的表达情况及其临床意义。方法:收集2008年10月至2009年7月,我院经病理证实的61例乳腺癌组织与相应癌旁组织标本以及26例乳腺良性肿瘤组织与相应瘤旁组织标本,分别通过实时PCR和Western印迹法在mRNA和蛋白质水平上检测KLK8在乳腺疾病中的表达情况。结果:KLK8在乳腺癌组织中的表达较其相应癌旁组织低(P<0.0001),在乳腺良性肿瘤中未发现该现象(P=0.424),并且其在正常乳腺癌组织中的表达比乳腺良性肿瘤组织中表达低(P=0.001)。KLK8的表达与乳腺癌的肿瘤大小、淋巴结转移情况及临床分期存在相关。结论:KLK8在乳腺癌中呈低表达,可能是乳腺癌预后良好的一个肿瘤生物学标志。
Objective: To investigate the expression of human kallikrein gene 8 (KLK8) in breast diseases and its clinical significance. Methods: From October 2008 to July 2009, 61 cases of breast cancer tissues and corresponding paracancerous tissues confirmed by pathology in our hospital and 26 cases of benign breast tumor tissues and corresponding tumor tissues were collected and analyzed by real-time PCR and Western Western blotting was used to detect the expression of KLK8 in breast disease at the mRNA and protein level. Results: The expression of KLK8 in breast cancer tissues was lower than that in its corresponding paracancerous tissues (P <0.0001). This phenomenon was not found in benign breast tumors (P = 0.424), and its expression was higher in breast cancer than in breast The expression of benign tumor tissue was low (P = 0.001). The expression of KLK8 correlates with the size of tumor, lymph node metastasis and clinical stage in breast cancer. Conclusion: The low expression of KLK8 in breast cancer may be a tumor biomarker with good prognosis in breast cancer.
