目的建立一种离子色谱-积分脉冲安培法测定燕窝、速食燕窝及银耳、海藻酸钠、鱼胶等常见伪品燕窝中N-乙酰神经氨酸的含量。方法采用Dionex ICS-5000离子色谱仪、Dionex ASRSRR500 4-mm抑制器、电导检测器测定,色谱柱为CarbonPac PAI柱,柱温为30℃,流动相为100 mmol/L NaOH+50 mmol/L NaAc,流速为1.0 mL/min,进样量为10μL,分别测定燕窝、速食燕窝及伪品中N-乙酰神经氨酸的含量。结果 N-乙酰神经氨酸在0.25~5.00μg/mL范围内线性良好,r=0.999 9;在燕窝中的平均回收率为99.7%,RSD为0.82%(n=6)。结论本方法灵敏度高、重复性好,可准确测定燕窝中N-乙酰神经氨酸的含量,为燕窝的质量控制提供了一种检测方法。
OBJECTIVE To establish a method for the determination of N-acetylneuraminic acid in bird’s nest, fast food bird’s nest and white fungus, sodium alginate and fish glue by ion chromatography-integrating pulse amperometry. The method was performed on a Dionex ICS-5000 ion chromatograph, a Dionex ASRSRR500 4-mm suppressor and a conductivity detector. The column was a CarbonPac PAI column with a mobile phase of 100 mmol / L NaOH + 50 mmol / L NaAc , The flow rate was 1.0 mL / min and the injection volume was 10 μL. The content of N-acetylneuraminic acid in the bird’s nest, fast food bird’s nest and counterfeit products were determined respectively. Results N-acetylneuraminic acid showed a good linearity in the range of 0.25-5.00 μg / mL with r = 0.999 9. The average recovery in the bird’s nest was 99.7% and the RSD was 0.82% (n = 6). Conclusion The method has high sensitivity and repeatability, and can accurately determine the content of N-acetylneuraminic acid in the bird’s nest, which provides a test method for the quality control of bird’s nest.
