目的:探讨针刺治疗CFS模式大鼠对其海马与下丘脑中Smad4蛋白表达的作用及其机制。方法:清洁级SD雄性大鼠48只,随机分为正常组、模型组、束缚组、针刺组,采用游泳多重应激刺激建立慢性疲劳综合征动物模型,针刺组选择百会、情感区、感觉区、给予电针治疗,采用Western blot技术检测各组Smad4蛋白表达水平;RT-PCR技术检测各组Smad4 mRNA表达水平。结果:western blot检测显示:CFS模型组大鼠海马与下丘脑中Smad4蛋白表达明显增多,而正常组则表达较低,针刺治疗后,针刺组大鼠海马与下丘脑中Smad4蛋白表达明显降低;RT-PCR法检测大显示:各组大鼠海马和下丘脑中均有Smad4mRNA表达,正常组Smad4 mRNA表达较弱,模型组Smad4 mRNA表达明显增加,治疗后,针刺组的Smad4mRNA表达水平明显降低。结论:电针百会、情感1区和感觉区能够降低CFS模型大鼠海马与下丘脑中Smad4 mRNA水平,从而抑制Smad4蛋白表达,进而对CFS疲劳状态起到改善和治疗作用。
Objective: To investigate the effect of acupuncture on the expression of Smad4 protein in hippocampus and hypothalamus of CFS model rats and its mechanism. Methods: Forty-eight male Sprague Dawley rats were randomly divided into normal group, model group, restraint group and acupuncture group. Animal model of chronic fatigue syndrome was established by swimming multiple stress stimulation. , Sensory area, given electroacupuncture treatment, Western blot was used to detect the expression of Smad4 protein in each group. The expression of Smad4 mRNA in each group was detected by RT-PCR. Results: The results of western blot showed that the expression of Smad4 protein in hippocampus and hypothalamus of CFS model group was significantly increased, while the expression of Smad4 protein in hippocampus and hypothalamus of acupuncture group was lower than that of normal group The mRNA and protein expression of Smad4 in hippocampus and hypothalamus of rats in each group were significantly lower than those in control group Obvious reduction. CONCLUSION: Electroacupuncture Baihui, Affective 1 and sensory areas can reduce Smad4 mRNA level in the hippocampus and hypothalamus of CFS model rats, thereby inhibiting the expression of Smad4 protein, thereby improving and treating CFS fatigue state.
