目的 :探讨 7 硝基吲唑 ( 7 NI)对缺氧缺血 (HI)新生大鼠脑组织c fos基因表达的影响。方法 :建立新生大鼠缺氧缺血性脑病(HIE)动物模型 ,采用RT PCR和免疫组化方法 ,检测 7 NI组 ,花生油 (PO)组 ,假手术 (SH)组及脑缺氧缺血 4组动物脑组织c fos基因表达。结果 :7 NI干预后 ,新生大鼠缺氧缺血性脑损伤 (HIBD)皮质、海马c fosmRNA于HI后 0~ 1h、Fos蛋白于HI后 2h明显下降(P <0 0 5 )。结论 :7 NI能抑制新生大鼠HIBD海马、皮质HI后 0~ 1hc fos基因和HI后 2hFos蛋白高表达。
Objective: To investigate the effect of 7-nitroindazole (7 NI) on c fos gene expression in brain tissue of hypoxic-ischemic (HI) neonatal rats. Methods: Animal models of hypoxic-ischemic encephalopathy (HIE) were established in neonatal rats. RT-PCR and immunohistochemistry were used to detect the effects of 7-NI, peanut oil (PO), sham-operation C fos gene expression in brain tissue of 4 groups. Results: After NIH intervention, c fos mRNA in cortex and hippocampus of hypoxic-ischemic brain damage (HIBD) rats was significantly decreased at 0-1h after HI and at 2h after HI (P <0.05). Conclusion: 7 NI can inhibit the expression of fos gene at 0 ~ 1hc and 2h after Fos HI in HIBD hippocampus and HI cortex in neonatal rats.
