目的:研究槲皮素对TNFα诱导的内皮细胞与中性粒细胞粘附的抑制作用及作用机制。方法:用髓过氧化酶法测定中性粒细胞与内皮细胞的粘附,ELISA法测定内皮细胞粘附分子的表达。结果:TNFα通过增加/诱导内皮细胞ICAM1,VCAM1及Eselectin的表达而剂量、时间依赖性地增加内皮细胞与中性粒细胞的粘附。槲皮素可剂量依赖性地抑制上述粘附,其作用机制为抑制TNFα诱导内皮细胞上述3种粘附分子的表达。结论:槲皮素通过抑制ICAM1,VCAM1及Eselectin的表达而降低TNFα诱导的内皮细胞与中性粒细胞的粘附。
Objective: To study the inhibitory effect of quercetin on TNFα-induced endothelial cell and neutrophil adhesion and its mechanism. Methods: The myeloperoxidase method was used to determine the adhesion of neutrophils to endothelial cells. The expression of endothelial cell adhesion molecules was measured by ELISA. RESULTS: TNFα increased the adhesion of endothelial cells to neutrophils in a time-and dose-dependent manner by increasing / inducing the expression of ICAM-1, VCAM-1 and E-selectin in endothelial cells. Quercetin inhibits the above adhesion in a dose-dependent manner, and its mechanism of action is to inhibit the TNFα-induced expression of the above three types of adhesion molecules in endothelial cells. Conclusion: Quercetin can reduce the TNFα-induced adhesion of endothelial cells to neutrophils by inhibiting the expression of ICAM-1, VCAM-1 and E-selectin.
