摘 要:目的:探讨在中小强度运动应激下右心室肌蛋白质组的差异表达及其规律,初步筛选与运动健康相关的备选目标蛋白质。方法: 将20只SD雄性大鼠随机分为对照组和实验组,以跑台运动的方式,复制递增运动负荷至24m·min-1的中小强度运动实验动物模型;提取右室肌组织的全蛋白进行双向凝胶电泳分离,运用ImageMaster 2D Platinum图像分析软件对2-DE胶进行分析。结果:2-DE图谱上,对照组检测蛋白质点437±17个,实验组检测448±19个;运动后47个蛋白质点发生了较明显的变化,其中,1个蛋白质点消失,表达量上调2倍以上的点20个,下调至50%以下的15个,上调在5倍以上的点8个,下调至20%以下的3个。结论:4周中小强度运动后,大鼠右心室肌蛋白质组发生了显著的变化,初步筛选出12个备选目标蛋白质点,为发现对研究运动健康与运动康复具有重要意义的新蛋白奠定前期工作基础。

The Influence of Four Weeks’ Medium Intensity Aerobic Exercise on Different Expression of Rat’s Right Ventricular Muscle Proteomics

Abstract: Objective: to explore the right ventricular proteomic’s different expression and regularity under the medium intensity exercise stress and to preliminary screen out the target proteins that related to exercise health. Methods: Twenty SD male rats were randomly divided into the control group and the exercise group, then the exercise group were trained on treadmill by copying the medium intensity exercise experimental animal models that workload increasing to 24 meters per minute, and then extracted the whole proteins from the right ventricular, using the technology of 2DE to separate the proteins, applying the image analysis software (ImageMaster 2D Platinum) to analysis the 2DE gels. Results: On the whole 2-DE map, the control group was detected 437±17 spots, the controlled group was detected 448±19 spots; after exercise, there were 47 protein spots changing obviously. In all of this protein spots, one spot disappeared; 20 spots increased more than 2 folds; 15 spots declined more than 50 %, 8 spots increased more than 5 folds and 3 spots declined to 20%.Conclusion: After four weeks’ medium intensity exercise, the proteomic of rats’ right ventricular muscle changed greatly, and preliminarily screened out 12 target protein spots, this protein spots will lay great foundation for discovering new proteins related to exercise rehabilitation and exercise health.
