目的:探讨土荆皮酸(PAB)和顺铂联用对人卵巢癌细胞株HO-8910裸鼠皮下移植瘤的生长及其血管生成的影响。方法:将20只荷人HO-8910卵巢癌的裸小鼠随机分成4组:①PAB组(60 mg/kg,腹腔注射治疗5天);②顺铂组(5 mg/kg,腹腔注射治疗5天);③联合用药组(PAB 60 mg/kg+顺铂5 mg/kg,腹腔注射治疗5天);④阴性对照组(给予同体积的含有6%聚乙二醇、3%乙醇、1%Tween 80的水溶液);计算瘤质量、瘤体积;同时应用CD34免疫组化染色,观测瘤组织内的微血管密度。结果:统计学分析表明:在抑瘤率和微血管密度方面,PAB组、顺铂组和阴性对照组比较差异具有统计学意义(P<0.01),联合用药组和其他3组比较差异均有统计学意义(P<0.01)。结论:PAB、顺铂对人卵巢癌移植瘤的生长具有抑制作用;PAB和顺铂联用能起增效作用。
OBJECTIVE: To investigate the effects of combination of Cephaloerythritol (PAB) and cisplatin on the growth and angiogenesis of subcutaneous xenografted human ovarian cancer cell line HO-8910 in nude mice. Methods: Twenty adult HO-8910 ovarian cancer bearing nude mice were randomly divided into 4 groups: ① PAB group (60 mg / kg, 5 days after intraperitoneal injection); ② cisplatin group (5 mg / kg, intraperitoneal injection of 5 Day); ③ combination group (PAB 60 mg / kg + cisplatin 5 mg / kg, intraperitoneal injection for 5 days); ④ negative control group (given the same volume containing 6% polyethylene glycol, Tween 80 aqueous solution); calculate the tumor mass, tumor volume; at the same time the application of CD34 immunohistochemical staining to observe the microvessel density within the tumor tissue. Results: Statistical analysis showed that there was significant difference between PAB group, cisplatin group and negative control group (P <0.01) in terms of tumor inhibition rate and microvessel density, there were statistical differences between the combination group and the other three groups Significance (P <0.01). Conclusion: PAB and cisplatin can inhibit the growth of human ovarian cancer xenografts. PAB combined with cisplatin can enhance the effect.
