目的对二氧化碳超临界萃取(CO2-SFE)和醇回流方法所提取的元胡白芷两种不同产物进行比较。方法采用固含物量、薄层色谱(TLC)、高效液相(HPLC)和不同的大鼠疼痛模型,从相关药效成分、提取物镇痛等角度对两种提取方法所得的产物进行比较。结果 CO2-SFE法所得萃取物的量约为醇回流法的1/4,纯度为后者的6~9倍;CO2-SFE法的提取物的镇痛作用强于醇回流法,且对血中疼痛物质的影响优于醇回流法。结论在元胡白芷的提取上,CO2-SFE明显优于醇回流技术,适宜进一步推广应用。
Objective To compare two different products of Angelica dahuricae extracted by carbon dioxide supercritical fluid extraction (CO2-SFE) and alcohol reflux method. Methods Solid content, thin layer chromatography (TLC), high performance liquid chromatography (HPLC) and different rat models of pain were used to compare the products obtained from the two extraction methods from the perspectives of the relevant pharmacologically active ingredient and the analgesic effect of the extract. Results The amount of extract obtained by CO2-SFE method was about 1/4 of the alcohol reflux method and the purity was 6 ~ 9 times of the latter. The extract of CO2-SFE had stronger analgesic effect than alcohol reflux method, The effect of pain substances is better than the alcohol reflux method. Conclusion In the extraction of Angelica dahurica, CO2-SFE is superior to alcohol reflux technology, which is suitable for further application.
