目的分离香鳞毛蕨Dryopteris fragrans的间苯三酚类化合物并探讨其抗真菌活性。方法采用硅胶柱色谱、Sephadex LH-20凝胶和制备HPLC等技术进行化学成分分离,根据理化性质和波谱数据鉴定化合物结构。采用美国临床实验室标准研究所(CLSI)制定的M38-A2方案(微量稀释法)研究化合物对红色毛癣菌和石膏样小孢子菌的抗真菌活性。结果从香鳞毛蕨中分离得到1对间苯三酚类同分异构体异黄绵马酸PB(1)、黄绵马酸PB(2)。化合物1和2对红色毛癣菌的最低抑菌浓度(MIC)分别为13.33μg/m L和1.875μg/m L,对石膏样小孢子菌的MIC分别为10μg/m L和1.325μg/m L。结论化合物1为新化合物,命名为异黄绵马酸PB。化合物2首次从该植物中分离得到。化合物1和2互为同分异构体,且对红色毛癣菌和石膏样小孢子菌均有较好的抑制作用。
OBJECTIVE To separate phloroglucinol from Dryopteris fragrans and investigate its antifungal activity. Methods The chemical constituents were isolated by silica gel column chromatography, Sephadex LH-20 gel and preparative HPLC. The structures of compounds were identified by their physico-chemical properties and spectral data. The antifungal activity of the compounds against Trichophyton rubrum and Plasmopara viticola was studied using the M38-A2 protocol (microdilution method) developed by the American Clinical Laboratory Standards Institute (CLSI). Results One pair of phloroglucinol isomers, isopumaranic acid PB (1) and xanthogen acid PB (2), were isolated from Fraxinus. The MICs of compounds 1 and 2 against Trichophyton rubrum were 13.33μg / m L and 1.875μg / m L, respectively, and the MICs were 10μg / m L and 1.325μg / m for G. gypseum respectively L. Conclusion Compound 1 is a new compound named as isocuproic acid PB. Compound 2 was isolated from this plant for the first time. Compounds 1 and 2 are isomers each other, and have a good inhibitory effect on Trichophyton rubrum and Gypsophila microsporum.
