目的 对河北汉族群体 6个短串联重复序列 ( short tandem repeat,STR)基因频率研究 ,以期得到河北汉族群体 D7S82 0、D19S2 5 3、D12 S391、D5 S818、D16 S5 39和 D8S1179基因座的群体遗传学数据。方法 随机抽取 173名河北省汉族人群无血缘关系个体的静脉血 ,应用 PCR技术扩增上述 6个短串联重复序列基因座 ,采用高分辨的聚丙烯酰胺凝胶垂直板电泳 ,银染显色分析。结果 得到了河北汉族这 6个位点的遗传多态分布。D7S82 0、D19S2 5 3、D12 S391、D5 S818、D16 S5 39、D8S1179基因座的杂合度分别为 :0 .82 8、0 .75 7、0 .76 9、0 .837、0 .785、0 .85 2。个体识别率分别为 :0 .914、0 .919、0 .94 0、0 .90 9、0 .917、0 .94 4。非父排除率分别为 :0 .6 18、0 .74 0、0 .80 1、0 .5 5 7、0 .6 5 5、0 .6 96。遗传多态性信息量分别为 :0 .771、0 .76 0、0 .76 2、0 .70 8、0 .776、0 .794。结论 上述 6个 STR基因座基因频率分布与 Hardy- Weinberg平衡吻合良好 ,在河北汉族群体中具有较高的非父排除率与个体识别率 ,在群体遗传学研究及法医学应用中有较高价值
Objective To study the frequency of six short tandem repeat (STR) genes in Hebei Han population in order to obtain the population heritability of D7S82 0, D19S2 5 3, D12 S391, D5 S818, D16 S5 39 and D8S1179 in Hebei Han population Learn data Methods A total of 173 venous blood samples of unrelated individuals from Han nationality population of Hebei Province were randomly selected. The six short tandem repeat loci were amplified by PCR and analyzed by high-resolution polyacrylamide gel vertical plate electrophoresis and silver staining . Results The genetic polymorphism distributions of these six loci in Hebei Han were obtained. The heterozygosity of D7S82 0, D19S2 5 3, D12 S391, D5 S818, D16 S5 39 and D8S1179 loci were: 0 .82 8,0 .75 7,0 .76 9,0 .837,0 .785,0 .85 2. Individual identification rates were: 0 .914,0 .919,0 .94 0,0 .90 9,0 .917,0 .94 4. Non-parent exclusion rates were: 0 .6 18,0 .74 0,0 .80 1,0 .5 5 7,0 .6 5 5,0 .6 96. The amount of genetic polymorphism information were: 0.771,0 .76 0,0 .76 2,0. 70 8,0 .776,0. 794. Conclusion The frequency distribution of the above 6 STR loci is in good agreement with Hardy-Weinberg equilibrium. In Han population of Hebei Province, there is a high non-father exclusion rate and individual recognition rate, which is of great value in population genetics and forensic applications
