目的:观察和解聚散方对单侧输尿管梗阻(UUO)致大鼠肾间质纤维化的结缔组织生长因子(CTGF)和层黏连蛋白(LN)的影响。方法:采用UUO建立大鼠肾间质纤维化模型,随机分为模型组、贝那普利组和和解聚散方组,给予相应药物灌胃,并设假手术组;UUO术后7天、14天和21天取材,采用免疫组化法检测各组手术侧肾组织CTGF和LN的表达。结果:与假手术组相比,UUO模型组7天、14天和21天的CTGF和LN的表达均增高(P<0.05);与模型组相比,和解聚散方组7天、14天和21天肾组织中的CTGF和LN表达均减少(P<0.05);21天和解聚散方组CTGF的表达比贝那普利组显著降低(P<0.05)。结论:和解聚散方可以降低肾间质纤维化不同时间点CTGF和LN的表达,表明了和解聚散方的有效性,验证了肾络聚散消长病机学说指导下的和解聚散法为延缓慢性肾脏病肾间质纤维化进展的有效治法。
OBJECTIVE: To observe the effect of reconciliation and polygation on connective tissue growth factor (CTGF) and laminin (LN) in rats with unilateral ureteral obstruction (UUO) induced by renal tubulointerstitial fibrosis. Methods: Rat model of renal interstitial fibrosis was established by UUO. The model rats were randomly divided into model group, benazepril group and reconciliation and dispersing group. The rats were given gavage and sham-operation group. After UUO for 7 days, 14th day and 21th day. The expression of CTGF and LN in each side of operation group were detected by immunohistochemistry. Results: Compared with the sham group, the expression of CTGF and LN in the UUO model group was significantly increased at 7, 14 and 21 days (P <0.05). Compared with the model group, (P <0.05). Compared with the benazepril group, the expression of CTGF and LN in 21 groups were significantly decreased (P <0.05). Conclusion: The reconciliation and polygation can reduce the expression of CTGF and LN at different time points of renal interstitial fibrosis, which shows the effectiveness of reconciliation and polygation.
