目的研究水疱性口炎病毒对胃肠道肿瘤细胞的选择性感染和杀伤作用。方法用0.1 pfu/细胞的水疱性口炎病毒分别感染胃癌细胞株(SGC-7901,BGC-823)、结肠癌细胞株(SW-480,HT-29)和脐静脉内皮细胞(HUVECs),应用标准噬斑测定技术滴定病毒在各类细胞中的产量,MTT法检测细胞增殖能力,TUNEL法检测细胞凋亡。结果 0.1 pfu/细胞的水疱性口炎病毒与上述5种细胞共同培养48h后,各细胞水疱性口炎病毒的产量分别为HUVECs(3.10±0.32)×104,SGC-7901(5.61±0.44)×105,BGC-823(6.3 2±0.36)×105,SW-480(7.22±0.82)×105,HT-29(6.97±0.18)×105。MTT结果显示除脐静脉内皮细胞外,其他细胞被大量杀死;TUNEL凋亡检测结果显示胃癌细胞和结肠癌细胞部分凋亡,脐静脉内皮细胞则很少凋亡。结论水疱性口炎病毒对上述胃癌细胞和结肠癌细胞有选择性感染和杀伤作用,并且能够诱导这些细胞凋亡,对正常细胞则影响不大,水疱性口炎病毒有希望成为有效的抗胃肠道肿瘤制剂。
Objective To study the selective infection and killing effect of vesicular stomatitis virus on gastrointestinal tumor cells. Methods Human gastric cancer cell lines (SGC-7901, BGC-823), colon cancer cell lines (SW-480, HT-29) and human umbilical vein endothelial cells (HUVECs) were infected with 0.1 pfu / cell vesicular stomatitis virus Standard plaque assay titration virus in a variety of cell yields, MTT assay cell proliferation, TUNEL assay apoptosis. Results The results showed that the rates of vesicular stomatitis virus (HUVECs) were 3.10 ± 0.32 × 104 and 5.61 ± 0.44 (HUVECs), respectively, after cultured with 0.1 pfu / cell vesicular stomatitis virus 105, BGC-823 (6.3 2 ± 0.36) × 105, SW-480 (7.22 ± 0.82) × 105 and HT-29 (6.97 ± 0.18) × 105. MTT results showed that except for umbilical vein endothelial cells, a large number of other cells were killed; TUNEL apoptosis test showed partial apoptosis of gastric cancer cells and colon cancer cells, but little apoptosis of umbilical vein endothelial cells. Conclusions The vesicular stomatitis virus has selective infection and killing effect on the above gastric cancer cells and colon cancer cells, and can induce the apoptosis of these cells with little effect on the normal cells. The vesicular stomatitis virus has the hope of becoming an effective anti-stomach Intestinal tumor preparations.
