目的探讨羧氨基葡聚多糖钠抑制胃癌脱落细胞种植转移的效果。方法取人胃癌MGC-803细胞株,分别加入12.5、25、50 mol/L羟氨基葡聚多糖钠,另设对照组加入生理盐水。采用MTT法检测胃癌细胞增殖情况,流式细胞仪检测细胞周期和凋亡情况,Transwell小室检测细胞迁移能力。结果羧氨基葡聚多糖钠显著抑制胃癌细胞的增殖,并呈剂量依赖性抑制(P<0.01);羧氨基葡聚多糖钠可引起胃癌细胞周期G1/S期阻滞和细胞凋亡,显著抑制胃癌细胞的转移(P均<0.01)。结论羧氨基葡聚多糖钠具有显著的抑制胃癌细胞生长及转移效应,可作为进展期胃癌根治性手术后腹腔冲洗液用于临床。
Objective To investigate the effect of carboxymethyl glucosaminoglycans on the growth and metastasis of exfoliative cells in gastric cancer. Methods Human gastric cancer cell line MGC-803 was obtained and added 12.5, 25 and 50 mol / L sodium glucosaminoglycans respectively. The control group was given normal saline. The proliferation of gastric cancer cells was detected by MTT assay. The cell cycle and apoptosis were detected by flow cytometry. Transwell chamber was used to detect cell migration. Results Sodium carboxymethyl glucosaminoglycans significantly inhibited the proliferation of gastric cancer cells in a dose-dependent manner (P <0.01). Sodium glucosaminoglycans induced cell cycle G1 / S arrest and apoptosis in gastric cancer cells and significantly inhibited Metastasis of gastric cancer cells (P <0.01). CONCLUSION Sodium carboxymethyl dextran can significantly inhibit the growth and metastasis of gastric cancer cells and can be used as a clinical postoperative peritoneal irrigation solution for advanced gastric cancer.
