目的探讨玻璃体腔注射曲安奈德治疗外伤性玻璃体出血的效果。方法建立20只外伤性玻璃体出血家兔模型。随机分为曲安奈德组和生理盐水组,每组各10眼。直接眼底镜观察玻璃体出血指数、增生性玻璃体视网膜病变(PVR)程度;ELISA测定玻璃体液中细胞因子浓度。结果曲安奈德组PVR程度、玻璃体腔白细胞介素-1β(IL-1β)、表皮生长因子(EGF)、转化生长因子β1(TGF-β1)浓度低于生理盐水组(P<0.05)。曲安奈德组玻璃体出血指数、玻璃体腔基质金属蛋白9(MMP-9)、血小板源性生长因子(PDGF)与生理盐水组比较差异无统计学意义。结论玻璃体腔注射曲安奈德可下调玻璃体腔IL-1β、TGF-β1、EGF表达,抑制增殖细胞的迁移和增生,从而防治PVR形成和发展。
Objective To investigate the effect of intravitreous injection of triamcinolone acetonide on traumatic vitreous hemorrhage. Methods Twenty rabbits with traumatic vitreous hemorrhage were established. Randomly divided into triamcinolone acetonide group and saline group, each group of 10 eyes. Direct ophthalmoscope vitreous hemorrhage index, proliferative vitreoretinopathy (PVR) degree; ELISA determination of cytokines in vitreous humor concentration. Results The levels of PVR, the levels of IL-1β, EGF and TGF-β1 in triamcinolone acetonide group were lower than those in saline group (P <0.05). Vitreous hemorrhage index, vitreous cavity matrix metalloproteinase 9 (MMP-9) and platelet-derived growth factor (PDGF) in triamcinolone acetonide group were not significantly different from those in saline group. Conclusion Intravitreal injection of triamcinolone acetonide can down-regulate the expression of IL-1β, TGF-β1 and EGF in the vitreous cavity and inhibit the migration and proliferation of proliferating cells, thus preventing the formation and development of PVR.
