本文对采用Sevage法去除水葫芦粗多糖中的粗蛋白进行了研究。试验采用单因素法,以试剂加入量、氯仿和正丁醇的比例、振摇时间等3个因素研究对粗蛋白去除率和多糖损失率的影响;再由单因素试验筛选的结果,设计了L_9(3~3)正交试验,试验结果表明:氯仿与正丁醇比例为4:1,试剂加入量为2:1,振摇时间15min为最佳工艺条件,除蛋白率为71.79%,多糖损失率为25.43%。
In this paper, Sevage method was used to remove crude protein in water hyacinth crude polysaccharide. The effect of crude protein removal rate and polysaccharide loss rate was studied by single factor method with three factors as the amount of reagents, the ratio of chloroform and n-butanol, shaking time and so on. The results of single factor screening showed that L9 (3 ~ 3) orthogonal experiment. The results showed that the optimum conditions were as follows: the ratio of chloroform to n-butanol was 4: 1, the amount of reagent was 2: 1 and the shaking time was 15min, the removal rate of protein was 71.79% The loss rate is 25.43%.
