研究了一种用于临床检测血清蛋白的毛细管区带电泳方法。弹性石英毛细管50 μm i.d.×47 cm (40 cm 有效长度),检测波长200 nm ,血清用运行缓冲液(含12.5 m m ol/L四硼酸钠、1 m m ol/L乳酸钙、0.7 m m ol/L硫酸镁,pH 9.70)稀释40倍,气压进样17.23 kPa·s,分析电压23 kV。正常血清蛋白分为6种,孕妇的分7种(多一个未知的α0峰)。将正常人、孕妇、多发性骨髓瘤和强直性脊柱炎患者的血清蛋白的毛细管电泳与传统的醋酸纤维膜电泳相比较,前者具有高分辨率、在线数据处理和自动化的特点。基于血清蛋白的诊断,毛细管电泳是一个可靠的分析新技术。
A capillary zone electrophoresis method for the clinical detection of serum proteins was studied. Flexible quartz capillary 50 μm i. d. × 47 cm (40 cm effective length) at a detection wavelength of 200 nm. Serum was eluted with running buffer (12.5 m mol / L sodium tetraborate, 1 m mol / L calcium lactate, 0.7 m mol / L magnesium sulfate, pH 9.70) was diluted 40 times, pressure was injected 17.23 kPa · s, the analysis voltage of 23 kV. Normal serum protein is divided into six kinds, pregnant women in seven (more than one unknown α0 peak). Capillary electrophoresis of serum proteins in normal, pregnant women, multiple myeloma, and patients with ankylosing spondylitis compared with conventional acetate-based membrane electrophoresis, the former with high-resolution, online data processing and automation features. Based on the diagnosis of serum proteins, capillary electrophoresis is a reliable new analytical technique.
