目的:通过高脂饮食诱导大鼠胰岛素抵抗(insulin resistance,IR)模型,观察抵当汤对其主动脉组织中胰岛素受体(insulin receptor,InsR)及胰岛素样生长因子1(insulin-like growth factor 1,IGF-1)以及血清中IGF-1的影响,探讨抵当汤改善胰岛素抵抗的作用机制。方法:雄性大鼠35只,随机分为5组,分别为:空白对照组、模型组、抵当汤低剂量组、抵当汤高剂量组、血脂康组。空白对照组喂普通饲料;模型组喂高脂饲料;抵当汤低、高剂量组喂高脂饲料,同时灌胃抵当汤煎剂1g/kg、2g/kg;血脂康组喂高脂饲料,同时灌胃血脂康0.2 g/kg,连续喂养8周。测定血清空腹血糖(FBG)、空腹胰岛素(FINS)和IGF-1,计算胰岛素敏感指数(ISI)和胰岛素抵抗指数(HOMA-IR);胸主动脉应用免疫组织化学方法,观察主动脉组织中InsR和IGF-1的变化。结果:抵当汤低、高剂量组与模型组相比,大鼠HOMA-IR明显降低,ISI明显升高(P<0.05或P<0.01);抵当汤低、高剂量组与模型组相比,大鼠平滑肌细胞胞膜及胞浆InsR表达较强,平均光密度(average optical density,AOD)统计学有显著性差异;抵当汤低、高剂量组与模型组相比,大鼠血清IGF-1明显升高(P<0.05);抵当汤低、高剂量组与模型组相比,大鼠平滑肌胞膜及胞浆IGF-1表达明显较强,AOD统计学具有显著性差异(P<0.05)。结论:抵当汤可降低IR大鼠HOMA-IR,升高ISI,提高胰岛素敏感性;增强IR大鼠平滑肌胞膜及胞浆中InsR表达,升高IR大鼠血清及主动脉组织中IGF-1水平,改善胰岛素抵抗。
OBJECTIVE: To observe the effects of dendang decoction on the expression of insulin receptor (InsR) and insulin-like growth factor 1 (insulin-like growth factor 1) , IGF-1) and IGF-1 in serum to explore the mechanism of action of Dan Dang Decoction in improving insulin resistance. Methods: Thirty-five male rats were randomly divided into five groups: control group, model group, low-dose group of Dangdang Decoction, high-dose group of Dangdang Decoction and Xuezhikang group. The blank control group was fed with normal diet; the model group was fed with high fat diet; the low and high dose group of Danzhintang was fed with high fat diet, while the rats were treated with dengtang decoction 1g / kg, 2g / kg; Xuezhikang fed with high fat diet, Oral Xuezhikang 0.2 g / kg, continuous feeding for 8 weeks. Serum fasting blood glucose (FBG), fasting insulin (FINS) and IGF-1 were measured and insulin sensitivity index (ISI) and insulin resistance index (HOMA-IR) were calculated. Thoracic aorta was used to observe the changes of InsR And IGF-1 changes. Results: Compared with the model group, the HOMA-IR of rats in the low and high dose groups of Doudang decoction decreased significantly and ISI increased significantly (P <0.05 or P <0.01) The mRNA expression of InsR in the membrane and cytoplasm of rat smooth muscle cells was stronger than that of the model group, and the average optical density (AOD) (P <0.05). Compared with the model group, the expression of IGF-1 in smooth muscle cells and cytoplasm of rats in low and high dose groups of Doudang decoction was significantly stronger (P <0.05) . Conclusion: Dendang Decoction can reduce HOMA-IR, increase ISI, increase insulin sensitivity in IR rats, enhance InsR expression in smooth muscle cells and cytoplasm of IR rats and increase IGF-1 in serum and aorta of IR rats Level, improve insulin resistance.
