目的观察Toll样受体4(toll-like receptor 4,TLR4)在结直肠癌和正常大肠组织中的表达情况,并且分析其表达与Fas配体(fas ligand,FasL)和表皮生长因子受体(epidermal growth factor receptor,EGFR)表达的关系。方法采用免疫组化SP三步法检测58例结直肠癌和25例正常大肠组织TLR4、FasL和EGFR的表达。通过Spearman’s相关分析,探讨TLR4与FasL或EGFR表达的关系。结果 58例结直肠癌中TLR4、FasL和EGFR表达的阳性率分别为59%、71%和83%,均显著高于其在正常大肠组织中的表达阳性率20%、22%和31%(P<0.05)。TLR4的表达与结直肠癌的远处转移具有相关性(χ2=4.063,P<0.05)。TLR4和FasL表达具有正相关性(r=0.249,P<0.01),TLR4表达强的部位多伴有FasL的强表达,TLR4表达弱的部位也常伴随FasL的弱表达。TLR4与EGFR的表达不具有相关性(r=0.345,P>0.05)。结论 TLR4的表达与结直肠癌远处转移有关,TLR4可能通过调控FasL的表达而参与结直肠癌的免疫反击。
Objective To observe the expression of toll-like receptor 4 (TLR4) in colorectal cancer and normal colorectal tissues and to analyze the relationship between its expression and Fas ligand (FasL) and epidermal growth factor receptor epidermal growth factor receptor, EGFR) expression. Methods The expressions of TLR4, FasL and EGFR in 58 cases of colorectal cancer and 25 cases of normal colorectal tissue were detected by SP immunohistochemical method. Spearman’s correlation analysis was used to explore the relationship between TLR4 and FasL or EGFR expression. Results The positive rates of TLR4, FasL and EGFR in 58 cases of colorectal cancer were 59%, 71% and 83%, respectively, which were significantly higher than those in normal colorectal tissues (20%, 22% and 31% P <0.05). The expression of TLR4 was correlated with the distant metastasis of colorectal cancer (χ2 = 4.063, P <0.05). There was a positive correlation between the expression of TLR4 and FasL (r = 0.249, P <0.01). The strong expression of TLR4 was accompanied by the strong expression of FasL. The weak expression of TLR4 was also accompanied by the weak expression of FasL. TLR4 and EGFR expression was not related (r = 0.345, P> 0.05). Conclusion The expression of TLR4 is related to the distant metastasis of colorectal cancer. TLR4 may be involved in immune counterattack of colorectal cancer by regulating the expression of FasL.
