目的研究胰腺癌患者血清miR-210的表达及意义,探讨血清miR-210与CA199联合检测在胰腺癌早期诊断中的价值。方法选取37例胰腺癌患者和40例健康对照者,应用实时荧光定量PCR检测血清中miR-210的表达水平;化学发光法检测血清中CA199含量;受试者曲线(ROC)分析miR-210、CA199及两者联合对胰腺癌的诊断价值。结果与对照组相比,胰腺癌患者血清中miR-210表达上调,CA199含量增加,差异均有统计学意义(P<0.05)。ROC分析结果显示,miR-210诊断胰腺癌的敏感度和特异度分别为80.9%和91.3%,均高于CA199(敏感度、特异度分别为63.2%、88.9%),miR-210和CA199联合检测的敏感度(95.5%)较单项指标明显提高。结论胰腺癌患者血清miR-210的表达上调,miR-210诊断胰腺癌的敏感度和特异度均高于CA199;两指标联合可明显提高诊断的敏感度。血清miR-210有潜力成为胰腺癌早期诊断的分子标志物之一。
Objective To study the expression and significance of serum miR-210 in patients with pancreatic cancer and to explore the value of combined detection of serum miR-210 and CA199 in the early diagnosis of pancreatic cancer. Methods Thirty-seven patients with pancreatic cancer and 40 healthy controls were enrolled in this study. The expression of miR-210 in serum was detected by real-time fluorescence quantitative PCR. The content of CA199 in serum was detected by chemiluminescence. The ROC curve was used to analyze the expression of miR- CA199 and their combined diagnosis of pancreatic cancer. Results Compared with the control group, the levels of miR-210 and CA199 in serum of pancreatic cancer patients were significantly increased (P <0.05). ROC analysis showed that the sensitivity and specificity of miR-210 in the diagnosis of pancreatic cancer were 80.9% and 91.3%, respectively, which were higher than those in CA199 (sensitivity and specificity were 63.2% and 88.9%, respectively). The combination of miR-210 and CA199 The detection sensitivity (95.5%) was significantly higher than the single index. Conclusions The expression of miR-210 in serum of patients with pancreatic cancer is up-regulated. The sensitivity and specificity of miR-210 in diagnosing pancreatic cancer are higher than those of CA199. The combination of two indexes can significantly improve the sensitivity of diagnosis. Serum miR-210 has the potential to become one of the molecular markers for the early diagnosis of pancreatic cancer.
