目的:检测维药西帕依溃结安对大鼠溃疡性结肠炎模型中诱导型一氧化氮合酶(iNOS)基因表达的影响,探讨药物的可能作用机制.方法:采用2,4-二硝基氯苯(DNCB)复合乙酸法构建溃疡性结肠炎大鼠模型,建立维药西帕依溃结安大、中、小剂量干预组及生理盐水阴性对照组(NS组),应用半定量RT-PCR和Western blot方法检测各组中iNOS mRNA和蛋白质的表达水平.结果:与NS组相比,维药西帕依溃结安各干预组iNOS mRNA表达水平无统计学意义,iNOS蛋白表达水平下调,差异有统计学意义(0.44±0.40vs1.00±0.07,P<0.05).结论:维药西帕依溃结安治疗溃疡性结肠炎可能是通过在转录后水平上调节iNOS的表达而发挥作用.
OBJECTIVE: To investigate the effect of xipaergujiean on the gene expression of inducible nitric oxide synthase (iNOS) in rat ulcerative colitis model and explore the possible mechanism.Methods: Nitrobenzene (DNCB) complex acetic acid method to establish ulcerative colitis rat model, the establishment of peacekeeping drug xipayu ulceration large, medium and small dose intervention group and saline negative control group (NS group), the application of semi-quantitative RT-PCR and Western blot were used to detect the expression of iNOS mRNA and protein in each group.Results: Compared with NS group, the expression of iNOS mRNA was not significantly different (0.44 ± 0.40vs1.00 ± 0.07, P <0.05) .Conclusion: The treatment of ulcerative colitis with paclitaxel may reduce the expression of iNOS at post-transcriptional level And play a role.
