补肾活血方对急性心肌梗死大鼠早期心功能及内皮素-1、血管紧张素Ⅱ的影响

目的观察补肾活血方对急性心肌梗死(AMI)大鼠早期心功能的影响并探讨其可能作用机制。方法将60只Wistar大鼠随机分为模型组、假手术组、阳性药组及补肾活血方高、中、低剂量组,每组10只。除假手术组外其余大鼠采用冠状动脉结扎法建立AMI大鼠模型。阳性药组予依那普利0.2 g/(kg·d)灌胃,补肾活血方高、中、低剂量组分别予补肾活血方生药10、5、2.5 g/(kg·d)灌胃,假手术组与模型组予等量生理盐水灌胃。14天后,采用高分辨率超声影像系统测量大鼠左心室舒张末期前壁厚度(LVAWd)与后壁厚度(LVPWd)、左室舒张末期内径(LVIDd)和收缩末期内径(LVIDs),计算左室舒张末期和收缩末期容积(LVEDV,LVESV)及射血分数(EF)、短轴缩短率(FS)、每搏输出量(SV),评价各组大鼠心脏结构和功能变化。ELISA法检测各组大鼠血清内皮素-1(ET-1)、血管紧张素Ⅱ(AngⅡ)水平。结果与模型组比较,补肾活血方各剂量组LVAWd均显著升高(P<0.05或P<0.01),LVIDs、LVIDd、LVEDV呈现减小趋势但差异无统计学意义(P>0.05);补肾活血方高剂量组与阳性药组EF、FS显著升高(P<0.01);补肾活血方各剂量组血清AngⅡ水平明显下降(P<0.01),ET-1水平呈现减小趋势但差异无统计学意义(P>0.05)。结论补肾活血方可在心肌梗死早期调节心室重构、改善心脏功能,其机制与调节Ang II、ET-1水平有关。 Objective To observe the effects of Bushen Huoxue Recipe (FQH) on early cardiac function in acute myocardial infarction (AMI) rats and to explore its possible mechanism. Methods Sixty Wistar rats were randomly divided into model group, sham operation group, positive drug group and Bushen Huoxue Fang high, medium and low dose group, 10 rats in each group. Except sham operation group, AMI rat model was established by coronary artery ligation. The rats in the positive group were given enalapril 0.2 g / (kg · d) orally and the rats in the high, medium and low dose of Bushen Huoxue prescription were given 10, 2.5 and 2.5 g / (kg · d) Sham operation group and model group were given the same amount of saline gavage. After 14 days, LVAWd and LVPWd, LVIDd and LVIDs of left ventricular end-diastolic pressure were measured by high-resolution ultrasound imaging system, (LVEDV, LVESV), ejection fraction (EF), short axis shortening (FS) and stroke volume (SV) were measured to evaluate the changes of cardiac structure and function in each group. Serum levels of endothelin-1 (ET-1) and angiotensin Ⅱ (AngⅡ) were detected by ELISA. Results Compared with the model group, the LVAWd of Bushen Huoxue prescription group was significantly increased (P <0.05 or P <0.01), LVIDs, LVIDd, LVEDV showed a decreasing trend but no significant difference (P> 0.05); Bushenhuoxue (P <0.01). The levels of serum AngⅡ in Bushenhuoxue Decoction group were significantly decreased (P <0.01), and ET-1 levels showed a decreasing trend but the difference was not statistically significant Significance (P> 0.05). Conclusion Bushenhuoxue can regulate ventricular remodeling and improve cardiac function in the early stage of myocardial infarction. Its mechanism is related to the regulation of Ang II and ET-1 levels.
