目的:探讨白介素-4(Interleukin-4,IL-4)基因589位点、白介素-4受体(interleukin-4receptor,IL-4R)基因576位点多态性与内蒙古地区汉族支气管哮喘患者是否存在遗传易感性,是否与血清总IgE浓度相关。方法:采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)方法检测内蒙古地区62例支气管哮喘患者和30例汉族正常人群IL-4基因的589位点、IL-4R基因的576位点多态性,进行基因型和基因频率分析,同时采用Elisa法检测患者血清总IgE浓度。结果:哮喘组IL-4基因启动子区-589(C/T)位点多态性分布频率与对照组比较,两组间基因型频率分析(X2=3.437,P=0.179),无显著性统计学差异(P>0.05);两组基因频率分析(X2=9.405,P=0.002),有显著性差异(P<0.05)。哮喘组IL-4R基因启动子区-576(Q/R)位点多态性分布频率与对照组比较,两组间基因型频率分析(X2=0.815,P=0.665),无显著性统计学差异(P>0.05),两组基因频率分析(X2=0.245,P=0.621),无显著性差异(P>0.05)。哮喘组血清总IgE浓度高于对照组,有显著性差异(t=6.367,P=0.00,P<0.05)。结论:内蒙古地区汉族人群哮喘组中,IL-4基因启动子区-589(C/T)位点多态性与支气管哮喘的发病无显著性差异;IL-4R基因-576(Q/R)位点多态性与支气管哮喘的发病无显著性差异;患者组血清总IgE显著高于对照组,但是与IL-4基因启动子区-589(C/T)位点多态性和IL-4R基因-576(Q/R)位点多态性没有相关性。
OBJECTIVE: To investigate the polymorphism of Interleukin-4 (IL-4) gene at 589, interleukin-4 receptor (IL-4R) gene 576 locus in Han nationality with bronchial asthma in Inner Mongolia Genetic susceptibility, whether with serum total IgE concentration. Methods: The 589 loci of IL-4 gene in 62 patients with bronchial asthma and 30 normal Han people in Inner Mongolia were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). IL-4R 576 loci polymorphism, genotype and gene frequency analysis, while using Elisa method to detect serum total IgE concentration. Results: Compared with the control group, the distribution frequency of -589 (C / T) polymorphism in IL-4 gene promoter region in asthma group was significantly lower than that in the control group (X2 = 3.437, P = 0.179) (P> 0.05). There was a significant difference (P <0.05) between two groups in gene frequency analysis (X2 = 9.405, P = 0.002). The distribution frequency of -576 (Q / R) locus of IL-4R gene promoter region in asthma group was significantly higher than that in control group (X2 = 0.815, P = 0.665) (P> 0.05). There was no significant difference (P> 0.05) between the two groups in gene frequency analysis (X2 = 0.245, P = 0.621). Serum total IgE levels in asthma group were significantly higher than those in control group (t = 6.367, P = 0.00, P <0.05). CONCLUSIONS: There was no significant difference in the polymorphism of IL-4 gene promoter -589 (C / T) locus and the incidence of bronchial asthma in the Han population of Inner Mongolia. IL-4R gene -576 (Q / R) There was no significant difference in the polymorphism of the IL-4 gene and the incidence of bronchial asthma. The serum total IgE of the patients was significantly higher than that of the control group, but not with the polymorphism of IL-4 gene -589 (C / T) 4R gene -576 (Q / R) locus polymorphism no correlation.
