目的探讨鱼藤酮对大鼠原代培养中脑细胞血红素加氧酶-1的影响。方法不同浓度的鱼藤酮处理原代中脑细胞培养不同时间,逆转录-聚合酶链反应(RT-PCR)和Western blot检测血红素加氧酶-1表达的改变。结果鱼藤酮可致神经元发生退变;血红素加氧酶-1表达水平,在鱼藤酮处理24h,0.01、0.1μmol/L浓度时增加显著,1.0μmol/L浓度组与对照组相比,差异无统计学意义;而1.0μmol/L鱼藤酮处理6h,血红素加氧酶-1表达显著升高,12和24h时无变化。而RT-PCR检测鱼藤酮处理24h后的血红素加氧酶-1表达水平无显著改变。结论鱼藤酮引起血红素加氧酶-1表达升高,可能与细胞受损程度有关。
Objective To investigate the effect of rotenone on heme oxygenase-1 in rat brain during primary culture. Methods Primary cultured rat brain cells were treated with different concentrations of rotenone for different time periods. The changes of heme oxygenase-1 expression were detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting. Results Rotenone induced neuronal degeneration. The expression level of heme oxygenase-1 increased significantly at the concentrations of 0.01, 0.01 and 0.1 μmol / L of rotenone for 24h, but no significant difference was found between 1.0 μmol / L and 1.0 μmol / L of rotenone Statistical significance; and 1.0μmol / L rotenone treatment 6h, heme oxygenase-1 expression was significantly increased, 12 and 24h did not change. RT-PCR detection of rotenone 24 h after heme oxygenase-1 expression levels did not change significantly. Conclusion Rotenone caused heme oxygenase-1 expression increased, which may be related to the degree of cell damage.
