本文介绍了应用PCR技术检测细胞培养和生物制品中支原体污染的研究进展。作者从标本处理、引物选择、反应条件和两步法PCR等方面论述了PCR检测支原体污染物的要点。与培养法和DNA荧光染色法相比,PCR具有高度敏感、特异、快速并能鉴定污染种别的优点。作者最后指出,在操作中,应严格执行实验室规范,以免产生假阳性结果。
This article describes the application of PCR technology to detect cell culture and biological products in the progress of mycoplasma contamination. The author discusses the main points of PCR detection of Mycoplasma contaminant from aspects of specimen processing, primer selection, reaction conditions and two-step PCR. PCR has the advantages of being highly sensitive, specific, rapid and capable of identifying contaminating species as compared to culture and DNA fluorescence staining. Finally, the author pointed out that in the operation, should strictly enforce the laboratory norms in order to avoid false positive results.
