目的:观察制首乌汤剂对阿霉素大鼠血液生化指标和肾组织的影响,探讨其对肾病综合征的防治作用及机制。方法:SD大鼠28只随机分为空白组、模型组、益肾通络方组和制首乌组,每组7只,对除空白组外的其余组建立阿霉素肾病模型。制首乌组、益肾通络方组分别灌服制首乌汤剂(0.21g/m L)和益肾通络方汤剂(2.00g/m L)治疗,空白组和模型组灌服蒸馏水,每天1次,连续3周。采用双缩脲法检测造模前、灌药前及灌药后24h尿蛋白;灌药后第8周各组大鼠眼眶后静脉丛取血检测血液生化指标,摘取肾脏进行肾组织病理学检查。结果:灌药前模型组及给药组24h尿蛋白含量均>200mg,提示造模成功;灌药后给药组含量均降低(P<0.01或P<0.05),益肾通络方组改善优于制首乌组(P<0.05)。给药组生化指标均优于模型组(P<0.01),益肾通络方组较制首乌组改善更显著(P<0.05)。病理学检查结果显示给药组均对肾小球损伤有一定的修复。结论:制首乌单独用于治疗肾病综合征有一定效果,复方应用效果更显著。
Objective: To observe the effects of Shouwu decoction on blood biochemical indexes and renal tissues in adriamycin-induced rats and to explore its preventive and therapeutic effects on nephrotic syndrome. Methods: Totally 28 Sprague-Dawley rats were randomly divided into blank group, model group, Yishen Tongluo Formula group and Shouyouwu group, with 7 rats in each group. Adriamycin nephropathy model was established in the rest except the blank group. Shouwu group and Yishen Tongluo recipe group were given Shouwu decoction (0.21g / m L) and Yishen Tonglu decoction (2.00g / m L) respectively. The blank group and model group were fed Distilled water once daily for 3 weeks. Urine protein was measured by biuret method before and 24 h after perfusion. Blood was collected from the orbital venous plexus at 8 weeks after perfusion to determine the blood biochemical parameters, and the kidneys were removed for renal histopathology an examination. Results: The 24-hour urinary protein content of the model group and the drug administration group were both> 200mg before irrigation, which suggested that the model was successful. The content of the drug group was decreased (P <0.01 or P <0.05), Yishen Tongluo Recipe It is superior to Shouwu group (P <0.05). The biochemical indexes in the treatment group were better than those in the model group (P <0.01), and the Yishen Tongluo Recipe group was significantly more effective than the Shouwu group (P <0.05). The results of pathological examination showed that the administration group had some repairs to glomerular injury. Conclusion: Radix Polygoni multiflori for the treatment of nephrotic syndrome alone has some effect, the compound application effect is more pronounced.
