目的观察软肝化坚颗粒对肝纤维化模型C57小鼠肝功能及转化生长因子β1(TGF-β1)的影响。方法将60只C57小鼠随机分为6组,即正常对照组、正常软肝化坚颗粒组、模型对照组、模型软肝化坚颗粒低剂量组、模型软肝化坚颗粒高剂量组和模型秋水仙碱组,每组10只。各模型组通过腹腔注射10%四氯化碳(CCl4)造模,造模2周后各用药组灌胃软肝化坚颗粒或秋水仙碱,用药6周时采集血清,测定小鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)及TGF-β1的含量。结果模型对照组小鼠血清ALT、AST及TGF-β1的含量均高于正常对照组(P<0.05),模型软肝化坚颗粒低剂量组、模型软肝化坚颗粒高剂量组以及模型秋水仙碱组小鼠血清ALT、AST的含量与正常对照组相比明显增高(P<0.05),正常软肝化坚颗粒组小鼠血清ALT含量与正常对照组相比降低(P<0.05),与模型对照组相比则明显降低(P<0.05),模型软肝化坚颗粒高剂量组血清ALT、AST水平低于模型秋水仙碱组(P<0.05)。模型软肝化坚颗粒组(包括低剂量和高剂量组)、模型秋水仙碱组和正常软肝化坚颗粒组小鼠血清TGF-β1的含量与模型对照组相比均明显降低,且均降低至正常对照组水平(P<0.05)。结论软肝化坚颗粒可以改善肝功能,抑制TGF-β1的产生,对实验性肝纤维化有一定的预防和治疗作用。
Objective To observe the effect of Ruanghujianjian Granule on liver function and TGF-β1 in C57 mice with hepatic fibrosis. Methods Sixty C57 mice were randomly divided into six groups: normal control group, normal soft liver capsule group, model control group, model soft liver and liver solid granules low dose group, model soft liver and liver solid granules high dose group Model colchicine group, 10 rats in each group. Each model group was established by intraperitoneal injection of CCl4 10%, and after 2 weeks of modeling, the rats in each group were given Rougan Huijian granules or colchicine, and the serum was collected 6 weeks after the treatment. The serum C Aminotransferase (ALT), aspartate aminotransferase (AST) and TGF-β1 content. Results The levels of serum ALT, AST and TGF-β1 in the model control group were significantly higher than those in the normal control group (P <0.05) The contents of serum ALT and AST in the mice treated with sodium bicarbonate were significantly higher than those in the normal control group (P <0.05). The serum levels of ALT in the mice treated with normal soft liver capsule were lower than those in the normal control group (P <0.05) Compared with the model control group, the levels of ALT and AST in model group were lower than those in model group (P <0.05). The contents of TGF-β1 in the models of Rougan Huijian granule group (including low-dose and high-dose groups), model colchicine group and normal soft liver capsule group were significantly lower than those in the model control group Reduced to normal control group (P <0.05). Conclusion Rougan Huajian Granule can improve liver function, inhibit the production of TGF-β1, and have certain preventive and therapeutic effects on experimental liver fibrosis.
