目的:研究淫羊藿女贞子配伍对哮喘大鼠模型Th1/Th2失衡的免疫调节作用。方法:采用OVA(ovalbumin,卵蛋白)致敏、激发复制大鼠慢性哮喘模型,以阿斯美胶囊为对照药,分别通过放免法及酶免法检测血及BALF(Bronchoalveolar lavage fluid,支气管肺泡灌洗液)中IL-4及IFN-γ的含量、免疫组化法检测肺组织IL-4和IFN-γ蛋白表达,RT-PCR技术检测肺组织T-bet和GATA-3 mRNA的表达,探讨淫羊藿女贞子对大鼠哮喘模型Th1/Th2调节途径的影响作用。结果:淫羊藿女贞子能显著抑制BALFIL-4含量(P<0.01),并降低血及BALF中IL-4/IFN-γ比值(P<0.05);对肺组织IL-4蛋白表达的Area有降低作用(P<0.05),并能抑制IL-4/IFN-γ Area的比值增高(P<0.01);上调肺组织T-bet mRNA表达,下调GATA-3 mRNA表达(P均<0.05)。结论:淫羊藿女贞子对哮喘大鼠Th1/Th2失衡有一定的调节作用。
Objective: To study the immunomodulatory effect of Epimedium sagittarius on imbalance of Th1 / Th2 in asthmatic rat model. Methods: Chronic asthma model was induced by ovalbumin (OVA) and replicated in asthmatic rats. Asimi capsule was used as control. Blood and BALF (Bronchoalveolar lavage fluid) were detected by radioimmunoassay and enzyme immunoassay. The contents of IL-4 and IFN-γ in the lung tissue were detected by immunohistochemistry. The expressions of T-bet and GATA-3 mRNA in lung tissue were detected by RT-PCR Effects of Epimedii Ligustrum lucidum on Th1 / Th2 Regulatory Pathway in Rat Asthma Model. Results: Ligustrum lucidum could significantly inhibit the content of BALFIL-4 (P <0.01) and decrease the ratio of IL-4 / IFN-γ in blood and BALF (P <0.05) (P <0.05) and increase the ratio of IL-4 / IFN-γ (P <0.01), upregulate the expression of T-bet mRNA and down-regulate the expression of GATA-3 mRNA in lung tissue ). Conclusion: Epimedii Ligustrum lucidum has a certain regulatory effect on the imbalance of Th1 / Th2 in asthmatic rats.
