目的探讨异丙酚对脓毒症大鼠肝线粒体呼吸功能的保护作用。方法 健康SD大鼠 18只,随机分为3组(n=6):假手术组(C组)、生理盐水组(NS组)、异丙酚组(P组)。NS、P组采用盲 肠结扎穿孔法(CLP)制备脓毒症模型,C组仅做开腹、分离盲肠远端、关腹手术。CLP后12 hP组颈外 静脉输注异丙酚5 mg/kg负荷量,继以10 mg·kg-1·h-1维持,C组、NS组输注生理盐水0.5 ml/h。持续 静脉输注异丙酚4 h时处死大鼠,取肝脏,采用Clark氧电极技术测定线粒体呼吸功能,分光光度法测 定丙二醛(MDA)、超氧化物歧化酶(SOD)、NO2/NO3-(NO)水平,采用定磷法测定ATP酶活性。结果 与C组比较,NS组、P组SOD及ATP的酶活性、呼吸控制率(RCR)、磷/氧比(ADP/O)降低,MDA水平、 态3及态4的呼吸速率升高(P<0.01);与NS组比较,P组SOD及ATP酶的活性、RCR及其ADP/O水 平升高,MDA、NO水平、态3及态4的呼吸速率降低(P<0.01)。结论 异丙酚通过中和自由基,对 脓毒症大鼠肝线粒体呼吸功能产生一定的保护作用。
Objective To investigate the protective effect of propofol on respiratory function of hepatic mitochondria in septic rats. Methods Eighteen healthy SD rats were randomly divided into three groups (n = 6): sham operation group (C group), saline group (NS group) and propofol group (P group). Sepsis model was made by cecal ligation and puncture (CLP) in NS and P groups. C group was only laparotomy, and the distal cecum and the abdomen were separated. After 12 h of CLP, propofol (5 mg / kg) was infused intravenously in the external jugular vein and then maintained at 10 mg · kg-1 · h-1. Rats in group C and NS were infused with saline 0.5 ml / h. Rats were sacrificed 4 h after intravenous infusion of propofol. The liver was taken and mitochondrial respiratory function was measured by Clark oxygen electrode. The contents of malondialdehyde (MDA), superoxide dismutase (SOD), NO2 / NO3 - (NO) levels, using the determination of ATP activity. Results Compared with group C, the activities of SOD and ATP, the respiratory control rate (RCR), the ratio of P / O (ADP / O) and the level of MDA and the respiration rate of state 3 and state 4 increased P <0.01). Compared with the NS group, the activities of SOD and ATPase, the levels of RCR and ADP / O in P group increased, the levels of MDA and NO, and the respiration rates in states 3 and 4 decreased (P <0. 01). Conclusion Propofol can protect liver mitochondria from sepsis rats by neutralizing free radicals.
