为了研究咪唑类离子液体在固定化纤维素酶降解小麦秸秆中的作用,试验采用小麦秸秆为原料,经离子液体预处理后,用壳聚糖固定的纤维素酶进行降解,计算产糖量。结果表明:0.25 g小麦秸秆(60目)用5 mL 1,3-二甲基咪唑磷酸二甲酯盐([Mmim]DMP)60℃超声处理30 min后,加入20 mL p H值4.8的柠檬酸缓冲液及固定化酶3.5 g,酶解温度为50℃,酶解时间为15 h,酶降解小麦秸秆产糖量最高,平均为2.76%。说明使用[Mmim]DMP预处理小麦秸秆可使固定化纤维素酶降解小麦秸秆的产糖量明显提高。
In order to study the effect of imidazole ionic liquids on the degradation of wheat straw by immobilized cellulase, wheat straw was used as the raw material. After pretreatment with ionic liquid, the cellulase degraded by chitosan was used to calculate the yield of sugar. The results showed that 0.25 mL wheat straw (60 mesh) was sonicated with 5 mL 1,3-dimethylimidazolium dimethyl phosphate ([Mmim] DMP) at 60 ℃ for 30 min, then 20 mL lemon Acid buffer and immobilized enzyme 3.5 g, enzymolysis temperature 50 ℃, enzymolysis time 15 h, enzymatic degradation of wheat straw sugar yield the highest, with an average of 2.76%. It shows that the pretreatment of wheat straw with [Mmim] DMP can significantly improve the yield of sugar by immobilized cellulase in wheat straw.
